摘要
目的探讨糖尿病并动脉粥样硬化兔血清中成纤维细胞生长因子(FGF)-21的表达及意义。方法12只新西兰兔按随机数字表分组。正常组(6只):耳缘静脉推注无菌生理盐水2 mL后用基础饲料喂养10周;糖尿病并动脉粥样硬化组(模型组6只),首先应用5%四氧嘧啶2 mL耳缘静脉推注复制糖尿病模型,然后行腹主动脉内膜球囊损伤术,术后用高脂饲料喂养10周。实验结束后禁食12 h以上,经耳缘静脉抽取空腹血离心,收集血清用酶联免疫法测FGF-21浓度,用氧化酶法测TC、TG、HDL-C和LDL-C浓度。处死后开腹取出腹主动脉固定,HE染色观察病理改变。结果模型组有明显的动脉粥样斑块形成;血清中FBG-21、TC、TG和LDL-C浓度均高于正常组,而HDL-C浓度低于正常组,差异均有统计学意义;多元线性逐步回归分析显示LDL-C是FGF-21的独立相关因素;二项Logistic回归分析显示,FGF-21与糖尿病并动脉粥样硬化相关。结论糖尿病并动脉粥样硬化兔血清中FGF-21浓度升高与脂类代谢紊乱有关。FGF-21可能与糖尿病并动脉粥样硬化有关。
【Objective】 To investigate the expression and its significance of fibroblast growth factor-21 in serum of diabetes combined atherosclerosis rabbits.【Methods】 12 New Zealand rabbits were randomly divided into two groups.In normal group,subjects were injected 2 mL N.S via ear vein and then use the basal diet for 10 weeks.In diabetes combined atherosclerosis group,subjects were injected 5% alloxan via ear vein and then theirs' abdominal aorta were injuried by balloon angioplasty,and used high fat diet for 10 weeks.After the experiment and fasted over 12 hour,the fasting blood were extracted via ear vein,then collect serum to measure FGF-21 concentration by ELISA,and measure TC,TG,HDL-C and LDL-C concentration by oxidase method.Abdominal aortic was fixed to observe the pathological changes by HE staining.【Results】 Diabetes combined atherosclerosis group had obvious atherosclerotic plaque formation.FBG-21,TC,TG and LDL-C concentration were higher than those of normal group,and HDL-C was lower than normal group,there has significant difference.Multiple linear stepwise regression analysis showed that LDL-C was the independent risk factor of FGF-21.Correlations were showed that FGF-21 with diabetes and atherosclerosis by logistic regression analysis.【Conclusion】 Increased FGF-21 concentrations and blood lipid metabolism disorder had related to diabetes combined atherosclerosis rabbits and maybe involved in FGF-21.
