摘要
目的探讨DJ-1/PTEN通路在糖尿病大鼠心肌缺血/再灌注期间对心肌损伤的影响。方法成年雄性SD大鼠,体质量220~280 g,随机分为3组:非糖尿病组(N组,n=10)、糖尿病假手术组(DM-S组,n=15)、糖尿病缺血/再灌注组(DM-IR组,n=15)。N组大鼠左锁骨中线开胸取心脏;DM-IR组采用结扎-开放左冠状动脉前降支(LAD)方法建立心肌缺血/再灌注模型,DM-S组只穿线包绕不结扎LAD。心肌缺血30min,再灌注120 min后处死。免疫组织化学法测定心肌组织DJ-1和PTEN的表达;光镜下观察心肌组织病理学变化;TTC法检测心肌梗死面积。结果与N组相比,DM-S组与DM-IR组DJ-1表达降低而PTEN表达升高(P〈0.05);与DM-S组相比,DM-IR组DJ-1表达降低而PTEN表达升高(P〈0.05);与DM-S组相比,DM-IR组心肌梗死面积明显增高(P〈0.05)。结论在糖尿病大鼠心肌及缺血/再灌注期间,DJ-1表达降低,进而对PTEN抑制作用减弱,从而导致心肌梗死面积增加。DJ-1/PTEN通路可能参与了糖尿病大鼠心肌病变及缺血/再灌注损伤的病理生理机制。
【Objective】 To investigate the effects of DJ-1/PTEN signal pathway in myocardial ischemia reperfusion on diabetes rats.【Methods】 Adult male SD rats weighing 220~280 g were used in this study.Type I diabetes mellitus was induced by STZ 60 mg/kg.The rats were randomly divided into 3 groups: nondiabetic group(Group N,n =10),diabetic sham-operated group(Group DM-S,n =15),diabetic ischemia/reperfusion group(Group DM-IR,n =15).In group DM-IR,ischemia-reperfusion model was induced by occlusion of LAD for 30 min and the occlusion of LAD was released for reperfusion 120 min.In group DM-S,LAD was exposed but not occluded.In group N,the hearts were removed rapidly.DJ-1,PTEN,pathological of Myocardial,TTC staining was measured.【Results】 Compared with group N,the expression of DJ-1 decreased,however,PTEN increased in other groups(P 0.05).Compared with group DM-S,the expression of DJ-1 decreased,while PTEN increased in group DM-IR(P 0.05),the myocardial infract areas increased in group DM-IR(P 0.05).【Conclusion】 The lower expression of DJ-1 can decrease the inhibition of PTEN,increase the myocardial infract areas.
