β-catenin及C-myc在胆总管囊肿中的表达及其意义

摘要 目的探讨β-catenin及C-myc在人胆总管囊肿中的表达情况。方法采用免疫组化方法检测成人胆总管囊肿患者(12例,成人组)、儿童胆总管囊肿患者(12例,儿童组)、足月新生儿尸体胆总管(12例,对照组)β-catenin和C-myc的表达情况。采用Western blot观察C-myc蛋白在胆总管囊肿中的表达情况。结果 (1)β-catenin表达在成人组、儿童组、对照组中分别为1 401.514 8±267.240 6、1 653.664 4±565.094 2、990.706 7±447.731 3,3组比较具有统计学差异(P=0.018 1);采用Dunnet检验显示:成人组、儿童组β-catenin表达均较对照组高,差异具有统计学意义(P<0.05),但成人组与儿童组比较差异无统计学意义(P>0.05)。(2)C-myc表达在成人组、儿童组、对照组中分别为800.837 6±378.740 6、1 822.167 3±1 301.414 4、933.537 3±494.420 7,3组间比较具有统计学差异(P=0.039 3);采用Dunnet检验显示:C-myc表达儿童组较对照组、成人组高(P值分别为0.028 2,0.007 9),但成人组和对照组比较无统计学差异(P>0.05)。Western blot检测结果显示,儿童组与对照组C-myc蛋白表达量分别为1.007 0±0.662 9、0.426 3±0.102 1,2组比较具有统计学差异(P=0.016 0)。结论β-catenin在儿童、成人胆总管囊肿中表达增高,C-myc在儿童胆总管囊肿中表达增高,β-catenin、C-myc可能参与儿童胆总管囊肿上皮的恶性转化。 Objective To investigate the expression of β-catenin and C-myc in human choledochal cyst(CCC).Methods The immunohistochemical method was used to detect β-catenin and C-myc expressions in choledochal cyst in different groups (group Ⅰ,adult patients with choledochal cyst,12 cases ;group Ⅱ,child patients with choledochal cyst,12 cases;group Ⅲ as control,term newborn corpses with normal common bile duct,12 cases).In addition,Western blot technique was adopted to further analyze C-myc expression in patients with choledochal cyst.Results The minimum qualifying scores(MQS) of β-catenin in adult CCC,children CCC and control groups were 1 401.514 8 ± 267.240 6,1 653.664 4 ± 565.094 2,and 990.706 7 ± 447.731 3,respectively.β-catenin expression in adult group and children group were higher than that in term newborn group,with significant difference of P =0.028 2 and P =0.007 9.The minimum qualifying scores (MQS) of C-myc in adult CCC,children CCC and control groups were 800.837 6 ± 378.740 6,1 822.167 3 ± 1 301.414 4,and 933.537 3 ± 494.420 7,respectively.C-myc expression in adult group and term newbom group were lower than that in children group.The significant difference between them were P =0.020 9 and P =0.030 3 respectively.For the C-myc results analyzed by Western blot,the protein expression in child patients with choledochal cyst was 1.007 0±0.662 9,and 0.426 3±0.102 1 in control group.There was a significant difference between the two groups (P =0.016 0).Conclusion The expression of C-myc was increased in children choledochal cyst,the expression of β-catenin was increased in children and adult choledochal cyst.β-catenin,C-myc might be associated with malignant transformation of choledochal cyst epithelium.
