摘要
目的观察依达拉奉对体外循环(CPB)大鼠脑损伤的影响。方法成年雄性SD大鼠24只,体重400-450g,采用随机数字表法,均分为三组:假手术组(S组)、CPB组(C组)和依达拉奉组(E组)。S组穿刺但不CPB;C组建立CPB模型;E组建立CPB模型并于预冲液中加入依达拉奉5mg/kg。于术后24h经股动脉抽血检测血清白细胞介素-6(IL-6)、IL-10、神经元特异性烯醇化酶(NSE)和S-100β蛋白含量;取脑组织匀浆,测丙二醛(MDA)含量和超氧化歧化酶(SOD)活性。结果与S组比较,C组和E组IL-6、IL-10、NSE、S-100β和MDA含量明显升高(P〈0.05),SOD活性明显降低(P〈0.05)。与C组比较,E组IL-6、NSE、S-100β和MDA含量明显降低,IL-10含量、SOD活性明显升高(P〈0.05)。结论预冲液中加入依达拉奉5mg/kg可明显减轻CPB大鼠脑的炎症反应和氧化应激反应,对CPB大鼠有一定的脑保护作用。
Objective To observe the effects of edaravone on cardiopulmonary bypass (CPB) induced brain injury in rats. Methods Twenty-four Sprague-Dawley male rats, weighing 400-450 g, were equally randomized into 3 groups (n=8) : sham operation group (group S), CPB group (group C) and edaravone group (group E). Rats in the group S were cannulated but did not undergo CPB. Rats in the groups C and E were subjected to CPB while the E group was also pretreated with edaravone 5 mg/kg in prime solutions. Blood samples were obtained via femoral artery 24 h after CPB to measure the serum concentrations of interleukin-6 ( IL-6 ), interleukin-10 ( IL-10 ), neuronespecificenolase (NSE) and S-10013 protein. The content of malondialdehyde (MDA) and the activity of superoxide dismutase (SOD) in brain tissue were also determined. Results Compared with the group S, the concentrations of IL-6, IL-10, MDA, NSE and S-100β protein increased while the activity of SOD decreased in the groups C and E (P〈0. 05). Compared with the group C, the concentrations of IL-6, MDA, NSE and S-100O protein decreased while IL-10 and SOD increased in group E (P 〈 0.05 ). Conclusion Edaravone 5 mg/kg pretreated in prime solutions can reduce inflammatory reaction and oxidative stress reaction, and exert protective effects on cardiopulmonary bypass induced brain injury in rats.
