异氟醚对大鼠S100、Tau和β淀粉样蛋白的影响 被引量:3

摘要 目的探讨异氟醚对成年大鼠S100、1、au和β淀粉样(Aβ)蛋白的影响。方法成年雄性SD大鼠36只,10周龄,体重250-280g,采用随机数字表法,将大鼠随机分为空白对照组(空气组,A组):大鼠不做任何处理;阳性对照组(氧气组,O组):大鼠于自制麻醉箱中吸入纯氧30min;麻醉组(T组):大鼠分别放入自制麻醉箱吸入异氟醚30min苏醒2h(T1组)、24h(T2组)、7d(T3组)和14d(T4组,每组6只)。采用酶联免疫吸附测定法测定血浆S100蛋白含量。分别在海马CA1、CA3区域测定Tau、Aβ蛋白阳性反应平均光密度。结果血浆S100蛋白A组为(211.5±105.4)pg/ml、O2组(197.2±81.9)pg/ml、T1组(142.6±42.4)pg/ml、T3组(214.6±90.2)和T4组(158.6±45.1)pg/ml,明显低于T2组(329.1±73.4)pg/ml(P〈0.05或P〈0.01)。与T2组比较,CA1区A组海马Tau蛋白阳性反应平均光密度明显增加(P〈0.05)。与T1组比较,CA3区A组、O组、T2组和T4组Aβ蛋白阳性反应平均光密度明显降低(P〈0.05或P〈0.01)。结论大鼠吸入异氟醚后S100、Tau、Aβ蛋白改变,可能导致大鼠一过性认知功能下降,随着时间的推移,学习记忆逐渐改善,不会对学习记忆造成永久性损害。 Objective To explore the changes of S100, Tau and Aβ after inhalation of isoflurane. Methods A total of thirty-six adult male Sprague-Dawley rats were randomized into 6 groups, control group(group A), positive control group(group O), anesthesia group 4 subgroups were divided as inhalation of domestic isoflurane, then recover 2 h(group T1), 24 h(group T2), 7d (group T3), 14 d(group T4), n= 6 respectively. At each time point after palinesthesia 1) took the orbital blood plasma S100 protein content; 2) detect the expression of hippocampal CA1 and CA3 of Tau and Aβ expression in the brain tissue. Results In the group T2, protein S100 was higher than that of the group A, T3(P〈0.05), much higher than that of the groups O, T1, T4(P〈0.01), Compared with the group A, the mean OD value in CA1 area of the group T2 increased(P〈0.05) ; the mean OD value in CA3 area has no statistical significance, Compared with the group A and T2, the mean OD value of T1 increased(P〈0. 01), compared with the group T4 and O, also increased (P〈0.01). Conclusion The changes of S100, Tau, Aβ caused by inhalation of isoflurane may be related to the short time learning and memory changes in SD rats.
