摘要
目的:观察人参皂苷Rg1的抗炎、镇痛作用并探讨其作用机制。方法:采用角叉菜胶致大鼠足肿胀模型观察人参皂苷Rg1的抗炎作用及对SOD活力和MDA含量的影响;采用RAW264.7细胞观察人参皂苷Rg1对脂多糖诱导的NO合成的影响;采用醋酸致小鼠扭体反应、福尔马林致大鼠疼痛反应两种模型观察人参皂苷Rg1的镇痛作用;采用原代培养的脊髓背根神经节神经细胞,使用荧光探针标记和膜片钳技术观察人参皂苷Rg1对辣椒素受体的影响。结果:人参皂苷Rg1可明显减轻角叉菜胶所致的大鼠足肿胀,降低大鼠致炎足中MDA含量,同时升高SOD活力;可抑制脂多糖诱导RAW264.7细胞产生NO;在醋酸致小鼠扭体反应、福尔马林致大鼠疼痛反应两种模型上镇痛效果良好;可抑制辣椒素受体激活产生的内向电流及Ca2+内流。结论:人参皂苷Rg1具有明显的抗炎、镇痛作用,抗氧化作用、抑制NO合成及抑制辣椒素受体激活可能是其发挥作用的机制。
OBIECTIVE To observe the anti-inflammatory and analgesic effects and the mechanism of actions of ginsenoside Rg. METHODS Models of carrageenan induced rats paw edema were performed to study the anti-inflammatory effects of gin senoside Rgl and its influences to SOD and MDA. The effect of ginsenoside Rgl on NO production induced by lipopolysaccha ride (LPS) in RAW264. 7 cells was observed. Methods of acetic acid-induced writhing in mice and formalin-induced pain in rats were Used to study the analgesic effect. Fluorescence probe and patch clamp technique were used to observe the effect of ginsen oside Rgl on transient receptor potential vanilloid i (TRPV1) in dorsal root ganglion (DRG) nerve cells. RESULTS Gineno- side Rgl significantly suppressed carrageen-induced rats paw edema, reduced MDA content and increased SOD activity. Ginenoside Rgl inhibited NO production induced by LPS in RAW264. 7 cells. The analgesic effect of ginenoside Rgl was ap patently observed in models of acetic acid-induced writhing in mice and formalin-induced pain in rats. The inward current and calcium influx induced by activation of TRPV1 were inhibited. CONCLUSION The anti-inflammatory and analgesic effects of ginenoside Rgl were obvious. Antioxygenation, inhibition of NO synthesis and activation of TRPVI may be related to its mechanism of actions
