摘要
通过研究Ru(II)多吡啶配合物[Ru(bpy)2(pmip)]2+与DNA的相互作用的光谱变化,发现[Ru(bpy)2(pmip)]2+-DNA体系相互作用模式既存在插入模式,也存在静电吸引模式。同时基于该体系增强的共振光散射现象,建立了一种简单、快速的测定纳克级核酸的新方法。在最佳反应条件下,体系在373 nm处共振光散射强度的增强与DNA的浓度呈线性关系,线性范围为0.065~2.400 mg/L,线性公式为△I RLS=144.09c+7.44(mg/L),相关系数为0.9985,ctDNA的检出限为6.04 ng/mL。
The interaction model in the system of [Ru(bpy)2(pmip) ]2+-DNA includes intercalation and electrostatic interaction through the study of spectral changes in the interaction between Ru(II) polypyridyl complexes [Ru(bpy)2(pmip) ]2+ and DNA system was found in this paper.A new method for the determination of nucleic acids at nanogram levels simply and rapidly based on the enhanced RLS intensity in the system was established.Under the optimal conditions,the enhanced intensity of RLS at 373 nm is proportional to the concentration of DNA.The linear range is 0.065 ~ 2.400 mg / L and the linear equation is △I RLS = 144.09c + 7.44(mg / L),the correlation coefficient(r) is 0.9985.The detection limit for calf thymus DNA is 6.04 ng / mL.Real samples were analyzed satisfactorily.
