摘要
目的建立榆树药材中咖啡酸的分析方法。方法以齐墩果酸对照品为对照,采用薄层层析(TLC)对榆树药材进行鉴别;采用高效液相色谱法(HPLC法)测定咖啡酸含量,色谱条件为:色谱柱:Phenomenex Luna C18(2)(4.6mm×150mm,5μm),流动相:甲醇一磷酸盐缓冲液(23:77),检测波长:323nm,流速:1.0ml/min,柱温:35℃。结果采自西藏林芝、山南地区的榆树药材4批,各批榆树药材TLC色谱中均能检出齐墩果酸;咖啡酸HPLC色谱峰与其他色谱峰分离良好,咖啡酸含量在0.03936~0.23616μg范围内,呈良好线性关系(r=0.9998),平均回收率为95.5%,RSD=1.16%。结论HPLC法简便快捷,结果可靠,为榆树药材的咖啡酸检测提供一个有效方法。
Objective To establish a method to determine the content of ulmus pumila in elm medicinal materi- als. Methods Thin Layer Chromatography(TLC) was adopted to identify the elm medicinal materials and High Performance Liquid Chromatography (HPLC)was adopted as well to determine elm medicinal material. The chromatographic condition were set: Phenomenex Luna C 18( 2 ), ( 4. 6 mm × 250 mm, 5μm ) as analytic col- umn ;moving phase as methanol- Phosphate buffer(23:77 ) ;detection wavelength at 323nm; column tempera- tore at 35℃ and flow rate at 1.0ml/min. Results Oleanic acid was identified in all the 4 batches of elm me- dicinal materials from Linzhi and Shannan by TLC method, and Caffeic acid was determined by HPLC. The lin- er range of contenf atalpol was 0. 03936 -0. 23616μg ,r = 0. 9998. The average recovery was 95.5%, RSD = 1.16%. Conclusion This HPLC method is characterized by simple, accurate, reproducible and can thus be employed to determine the caffeic acid content in ulmus pumila.
