形寒与寒饮对小鼠肺组织水通道蛋白-4表达的影响及意义 被引量:4

摘要 目的:观察形寒与寒饮对小鼠肺组织水通道蛋白4(AQP4)表达的意义。方法:40只昆明小鼠随机分为4组:常温对照组、形寒组、寒饮组、形寒寒饮组,每组1 0只,置于人工气候箱处理后取小鼠肺脏进行病理切片,Western blot法检测小鼠肺组织中AQP-4蛋白的表达情况。结果:形寒组、寒饮组、形寒寒饮组与常温对照组比较,病变程度以形寒寒饮组尤为严重。形寒寒饮组小鼠肺部出现大量的淋巴结样的结构和脓肿样的结构病变,形寒组见到大量的炎细胞浸润,寒饮组较少的炎性病变。肺组织AQP4蛋白的表达值亦均下降,以形寒寒饮组下降最明显,形寒组次之,寒饮组再次之,差异具有统计学意义,组间AQP4蛋白值差异均有统计学的意义。结论:实验证明了单纯的"形寒"与"寒饮"亦能造成肺脏的损伤,以形寒寒饮两者合并时对肺脏的损伤程度最为严重,提示形寒、寒饮等致病因素可导致肺水肿与肺组织AQP4蛋白表达下降相关,以形寒寒饮两者相合尤为明显。 Objective : To investigate the effects of aquaporin 4 ( AQP4 ) expression of mice body cold and C^ld fluid retention. Methods : 40 Kunming mice were divided into four groups : normal group, body cold group, cold fluid retention group and body cold and cold fliud retention group; 10 for each group.The mice were put into different temperature-controlled boxes or given different stimulations.During this period, their changes, such as their skin, fur and behaviors were observed.The samples were collected to observe their pathological changes in pulmonary tissue and AQP5 water channel protein expression with Western-blot after 15 days. Results : Compared with normal group, Body cold and cold fluid retention group had the most serious changes.There were a lot of lymph node-like structures and abscess-like structures in lung tissuses of mice in body cold and cold fluid retention group.There were lots of inflammation ceils in body cold group while in cold fliud retention group there were few.AQP4 protein expression values were decreased in all groups, expecially in body cold and cold fluid retention group, followed by body cold group, and then cold fluid retention group.The difference was statistical significant ( P〈0.01 ). Conclusion : Experiments show that the simple-body cold and cold fluid retention can cause lung damage.The degree of injury is the most serious in body cold and cold fluid retention group.It prompted body cold, cold fluid retention and other risks may lead to pulmonary edema and lung tissue.AQP4 protein expression associated with a decrease, escepically body cold and cold fluid retention.
