摘要
目的:观察外感湿邪对胶原诱导关节炎(CIA)大鼠血清CD44水平的影响。方法:选取Wistar大鼠50只,随机分成5组,每组10只,分别为正常组、模型组、风寒湿组、风湿组及风湿热组,造模1个月后取大鼠踝关节滑膜进行病理切片,同时摘取眼球取血,用ELISA检测血清中CD44的变化情况。结果:模型组、风寒湿组、风湿组及风湿热组与正常组比较,滑膜细胞都有不同程度的增生,以风寒湿组增生最为明显,其次为风湿热组,风湿组次之,模型组增生程度最低;血清CD44水平均亦明显升高,差异有显著意义;5个实验组两两比较CD44值差异均有统计学意义。且各组血清CD44水平与其滑膜炎症程度呈正相关。结论:风、寒、湿、热等外邪对CIA发病是有影响的,影响程度以风寒湿组最为严重;在各造模组中,CD44水平明显升高,提示CD44可能参与了CIA的发生,且风、寒、湿、热等外在因素能加重CD44的表达,其中风寒湿组CD44表达值最高。
Objective: To observe the effect of the invasion of wind cold damp and heat evil on CD44 in collagen induced arthritis ( CIA ) rats. Methods : In this research, the natural damp environment was simulated.50 Wistar rats were randomly divided into 5 groups: the normal group, model group, the wind cold damp ( WCD ) group, wind damp ( WD ) group and wind damp heat ( WDH ) group, each group only 10.The ankle synovial was taken out for pathological section at day 30 after experiment.The concentration of CD44 in serum was measured by enzyme-linked immunosorbent assay. Results: Compared with normal group, the levels of CD44 in model group, the wind cold damp group, wind damp group and wind damp heat group were obviously higher in the serum ( P〈0.01 ) .The increase in the wind cold damp group was the most obvious, followed by wind damp heat group, wind damp group was the second, and model group was the minimum ( P〈0.01 ) .And there was a positive correlation between the level of serum CD44 and synovial hyperplasia. Conclusion : The invasion of wind cold damp and heat evil has an effect on the happening of CIA, and a^gravates the expression of CD44.
