摘要
目的检测白藜芦醇作用MCF-7(野生型p53)和MDA-MB-231(突变型p53)乳腺癌细胞后ASPP1和ASPP2的表达。方法采用MTT法检测白藜芦醇对乳腺癌细胞增殖的影响,采用Western blot法检测经白藜芦醇处理后,MCF-7、MDA-MB-231和MCF-7/Caspase3细胞株PARP的蛋白水平变化。采用RT-PCR、Western blot法检测经白藜芦醇处理后,MCF-7、MDA-MB-231细胞中ASPP1和ASPP2的mRNA和蛋白表达变化。结果白藜芦醇对乳腺癌细胞MCF-7和MDA-MB231的生长均有抑制作用,呈浓度依赖性。MCF-7和MDA-MB-231细胞中ASPP1和ASPP2的mRNA和蛋白表达均较低,与MDA-MB-231相比,MCF-7细胞中ASPP2的蛋白表达更低。白藜芦醇作用于细胞后,与对照组相比,ASPP1和ASPP2的mRNA和蛋白表达均升高,且随药物浓度增高而表达增高;ASPP1和ASPP2的mRNA和蛋白表达与肿瘤细胞的凋亡呈正相关。结论白藜芦醇诱导乳腺癌细胞ASPP1、ASPP2基因高表达。
Purpose To examine the expression of ASPP1 and ASPP2 in MCF-7 (wild-type p53 ) and MDA-MB-231 (mutant-type p53 ) cells treated with resveratrol. Methods MTT assay was used to evaluate the growth inhibition of MCF-7 and MDA-MB-231 cells in responses to various doses of resveratrol. Western blot was applied to detect the expression of PARP protein levels in MCF-7, MDA- MB-231 and MCF-7/Caspase3 treated cells with resveratrol. RT-PCR, Western blot were applied to detect the expression of ASPP1 and ASPP2 mRNA and protein levels in MCF-7 and MDA-MB-231 cells treated with resveratrol. Results Proliferation of MCF-7 and MDA-MB-231 cells was inhibited by resveratrol in a dose dependent manner. The expression of ASPP1 and ASPP2 mRNA and protein in MCF-7 and MDA-MB-231 cells treated with resveratrol were increased in a dose-dependent manner as determined by RT-PCR, Western blot analysis. Conclusion Resveratrol induced apoptosis correlates with induction of ASPP1 and ASPP2 expression.
