摘要
目的通过实验观察茶黄素和脑血通口服液对动脉粥样硬化(AS)大鼠主动脉细胞结构、血脂和血液流变学指标的影响,探讨茶黄素和脑血通口服液治疗的作用机制。方法将80只Wistar大鼠随机均分为:①正常对照组:未造模,喂饲普通饲料,灌胃生理盐水1 ml/100 g·d;②AS模型组:造模成功的Wistar大鼠,灌胃生理盐水1 ml/100 g·d;③茶黄素组:造模成功的Wistar大鼠,每天灌胃茶黄素药液2.125 g/kg,蒸馏水稀释为浓度1.5 g/ml;④脑血通口服液组:造模成功的Wistar大鼠,每天灌胃脑血通口服2.125g/kg,蒸馏水稀释为浓度2.5 g/ml。茶黄素组和脑血通口服液组Wistar大鼠每日灌胃药液同时高脂饲料喂养,连续给药60 d,AS模型组灌胃等体积蒸馏水。对各组进行血液流变学和血脂指标进行分析,并观察AS大鼠主动脉细胞结构的变化。结果茶黄素组与AS模型组血脂比较有明显改善作用;扫描电子显微镜观察茶黄素组大鼠主动脉根部细胞较光滑呈修复状态,细胞间较有规则与方向性排列,细胞间有少量沉积较紧密连接。结论茶黄素对AS大鼠血脂和血液流变学指标可以进行调节,有降低血液黏稠度作用,促进受损细胞修复作用。
Objective Experimental observation on the effects of aortic cell structure, lipid and hemorheology of atherosclerosis rats, that treated by theaflavins and naoxuetong granule, in order to explore the corresponding mechanisms. Methods 80 Wistar rats were randomly divided, (1) normal control group: no modeling, fed with normal diet, given normal saline 1 ml/100 g · d; (2) atherosclerosis model group: successful modeling using Wistar rats, given normal saline 1 mE/100 g ·d; (3) theaflavins group: successful modeling using Wistar rats, daily detured theaflavins liquid 2. 125 g/kg, diluting the concentration of 1.5 g/ml by distilled water; (4) naoxuetong granule group: successful modeling using Wistar rats, daily orally detured by 2. 125 g/kg, diluting the concentration 2.5 g/ml by distilled water. Among them, the rats treated with theaflavins and naoxuetong granule were fed daily stomach with fat diet simultaneously , continuous administration of 60 d. The model group was given an equal volume of distilled water. Rheology of each group and serum lipids were analyzed and observed, with the rat aortic atherosclerotic changes in cell structure. Results In contrast to the model group, the theaflavins group was significantly improved on the blood lipid level. Scanning electron microscopy results revealed that, treated by theaflavins, the aortic root cells were relatively smooth repair status; cells were more rules and orientation, during the deposition of a small amount of cells, there exist more close connections. Conclusion Theaflavins on atherosclerotic lipid and hemorheology in rats can be adjusted, having lower blood viscosity role in promoting the repair of cells.
