rmCT-1对坐骨神经再支配骨骼肌形态和功能影响

EFFECT OF RECOMBINATION MOUSE CARDIOTROPHIN-1ON MORPHOLOGY AND FUNCTIONAL RECOVERY OF THE REINNERVATED SKELETAL MUSCLE

摘要 目的探讨重组小鼠心脏营养素-1(rmCT-1)对坐骨神经再支配骨骼肌形态和功能的影响。方法成年Swiss小鼠60只,切断右侧坐骨神经并立即修复,随机分为实验组和对照组,每组30只,术后每天分别腹腔注射rmCT-1(100μg/kg)及等量CT~1溶媒,4、8和12周后测量坐骨神经再支配骨骼肌腓肠肌湿质量、肌纤维截面积和总蛋白含量、复合肌肉动作电位波幅(CMAP)、肌张力、再生坐骨神经有髓神经纤维数,并与对照组比较。结果与对照组相比,实验组小鼠各时间点再支配骨骼肌腓肠肌湿质量、肌纤维截面积和总蛋白含量、CMAP、肌张力以及再生坐骨神经有髓神经纤维数均有显著提高,差异有显著性(t=6.39~82.53,P〈O.01)。结论rmCT-1可以明显促进坐骨神经再支配骨骼肌的形态和功能的恢复。 Objective To investigate the morphology and functional recovery of reinnervated skeletal muscle treated with recombination mouse cardiotrophin-1 (rmCT-1). Methods Sixty Swiss mice underwent a right-sided sciatic nerve disconnection and immediate restoration, and then equally divided into experimental group and control group. After surgery, daily intraperitoneal injection of rmCT-1 (100 μg/kg) and same dose of CT-1 solvent were offered, respectively. The wet weight of skeletal muscle, cross-sectional area, total protein, CMAP, the fibre number of the repaired sciatic nerve and latency and tension of reinnervated skeleton muscle were assayed 4, 8 and 12 weeks after surgery. Results Compared with the control,the number of wet weight of skeletal muscle, cross-sectional area, total protein,CMAP, the fibre number of the repaired sciatic nerve and latency and tension of reinnervated skeleton muscle of the mice treated with rmCT-1 were all significantly increased (t=6.39-82.53,P〈0.01). Con- clusion RmCT-1 can effectively improve the morphological and functional recovery of the reinnervated skeletal muscle.
