中国番茄黄瓜花叶病毒血清组的鉴定 被引量:10

The Identification of Subgroup of CMV from Tomato in China by DAS-ELISA

摘要 来自我国番茄主产区的 58个黄瓜花叶病毒 (CMV)分离物包含有轻花叶株系、重花叶株系、坏死株系和黄化株系。用分别代表CMV亚组Ⅰ和CMV亚组Ⅱ的两个标准毒株CMV -D和CMV -S的抗血清进行DAS -ELISA鉴定 ,有 56个CMV分离物与CMV -D的抗血清起强烈的阳性反应 ,划归为CMV亚组Ⅰ ,占CMV分离物总数的 96.6% ;有两个CMV分离物与CMV -S的抗血清起强烈的阳性反应 ,划归为CMV亚组Ⅱ ,占CMV分离物总数的 3.4 % ,这是首次从侵染我国番茄CMV中鉴定出属于CMV亚组Ⅱ的分离物。属于CMV亚组Ⅱ的两个分离物 (GD - 14和HN - 11)均为黄化株系。 Fifty eight cucumber mosaic virus(CMV)isolates from tomato in China,belonged to different strains:i.e.,mild mosaic,severe mosaic,necrotic and yellow strain of CMV,were identified with antibodies to CMV-D and CMV-S respectively by double antibodies sandwich enzyme linked immunosorbent assay(DAS-ELISA).The CMV-D and CMV-S strains were two standard ones which represented subgroup Ⅰ and Ⅱ of CMV respectively.Fifty six CMV isolates showed the strong positive reaction with antibody to CMV D,should belong to CMV subgroup Ⅰ and occupied 96.6% of total CMV isolates.Two CMV isolates(GD-14 and HN-11)showed the strong positive reaction with antibody to CMV-S,should belong to CMV subgroup Ⅱ and made up a lower percentage at 3.4%. It was the first report to confirm the existence of the CMV subgroup Ⅱ of tomato in China.The GD-14 and HN-11 isolates belonged to yellow strain in our study.Whether or not they contain the satellite RNA is a problem being worthy of research.
