摘要
取河套蜜瓜 (Cucumis melo L .cv Hetao)在 MS培养基上生长至 5 d的幼嫩子叶 ,在 MS+ 6 - BA0 .5 +IAA1 .0 的培养基中直接诱导生芽 ,待再生芽长至 0 .4cm~ 0 .6 cm时 ,切下转入芽伸长培养基中 (MS+ 6 - BA0 .9) ,待芽伸长至 0 .8cm~ 1.2 cm时 ,再转入生根诱导 (MS+ IAA2 )中 ,待植株长至 8cm~ 9cm时 ,转入沙质土壤中炼苗。整个过程约需 6 5 d~ 75 d,单个外植体诱导成芽最多达 31个 ,再生植株诱导率可达 6 2 .5 % ,为河套蜜瓜的快速繁殖和遗传转化提供了良好的再生系统。
The Cotyledon explants of Cucumis melo L.cv Hetao were excised after 5days of germination on MS medium followed by transfer into the shoot indcuing medium (MS+6-BA0.5ml/L+IAA1.0mg/L)for shoot formation.First shoots were formed after 15 days.The generated shoots(0.4~0.6cm in height) were cut and transfered into shoot lengthening from 0.8cm to 1.2cm,then transfered into rooting medium.The generated shoots (1.2cm in height) were transplanted into sandy soil pots for intact plant formation.The total process of regeneration took 65~75 days.The average regenerated rate is 62.5%.
