摘要
选用 8 0个随机引物 ,对栽培种陆地棉、野生种雷蒙德氏棉、它们的杂种一代以及从回交后代中选育的 9个种质系进行了RAPD指纹图谱和遗传相似性研究。结果表明 :2 9个随机引物扩增的带具多态性 ,并能将两个不同棉种、F1 和种质系相互间加以区别。在 9个种质系中分别检测到 5~ 9条 (3 .7%~ 5 .3 % )带与野生种相同的特异带 ,大多数带与陆地棉回交亲本一致。野生种与种质系间的相似系数为 0 .388~ 0 .479,陆地棉亲本与种质系间相似系数为 0 .6 36~ 0 .892 ,材料间的带型差异与相似系数是吻合的。因此 ,利用RAPD标记检测远缘杂种后代材料中来自野生种的血缘是可行的。
RAPD(random amplified polymorphic DNA) markers generated by 80 random primers were used to fingerprint and assess genetic similarity of nine germplasm lines derived from ( G. hirsutum×G. raimondii ) backcrossing generation, parents and F 1. The result showed that 29 primers were polymorphic among upland cotton, G.raimondii , F 1 and germplasm lines. There were 5~9 bands in germplasm lines identical to wild species. Most bands of germplasm lines are the same as upland cotton parents. The similarity coefficients between wild species and its filial lines were 0.388~0.479, those similarity between upland cotton and germplasm lines were 0.636~0.892, vice verse. The consistency between DNA ploymorphic and similarity of the materials showed that RAPD markers are reliable for determining genetic variation among accessions derived from interspecific hybrid.
