半夏泻心汤水溶提取物治疗BALB/c小鼠恶唑酮结肠炎的疗效及机制 被引量:6

Efficacy and Mechanism of the Water-Soluble Extracts of Banxiaxiexin Decotion in Oxazolone-Colitis in BALB/c Mice

摘要 目的:探讨半夏泻心汤水溶提取物对BALB/c小鼠恶唑酮结肠炎的疗效及机制。方法:雄性BALB/c小鼠40只,体质量18~20 g,其中30只第1天和第2天用3%恶唑酮溶液200μL皮肤致敏,第6天用1%恶唑酮溶液150μL保留灌肠,建立BALB/c小鼠恶唑酮溃疡性结肠炎模型。对照组10只,分别给予等体积、等浓度的乙醇涂擦皮肤、灌肠。30只小鼠造模成功后分为模型组、柳氮磺胺吡啶(salicylazosulfapyridine,SASP)组、半夏泻心汤组(简称半夏组),每组各10只。半夏组采用半夏泻心汤水溶提取物以10 mg/g(体质量)灌胃治疗7 d;SASP组采用SASP以20 mg/g(体质量)灌胃治疗7 d;模型组和对照组小鼠给予等体积水灌胃;每天1次,共7 d。7 d后,对各组小鼠进行疾病活动指数(disease activity index,DAI)评分、组织病理学炎性反应评分,并采用荧光定量逆转录聚合酶链反应方法测定结肠组织白细胞介素-5(IL-5)mRNA和白细胞介素-13(IL-13)mRNA的表达。结果:模型组小鼠的DAI评分(1.6±0.3)和组织病理学炎性反应评分(2.7±0.7)均显著高于对照组(0.2±0.1,1.7±0.5),而且模型组结肠组织IL-5 mRNA和IL-13 mRNA的表达(50.897±23.260,155.009±76.642)亦显著高于对照组(1.891±1.281,4.483±3.884)。半夏泻心汤水溶提取物能显著降低DAI评分和组织病理学炎性反应评分(0.3±0.3,1.5±0.4),而且结肠组织IL-5 mRNA和IL-13 mRNA的表达(7.822±5.321,39.999±22.420)亦显著降低,其疗效与SASP相当(0.4±0.2、1.3±0.6;21.297±20.800、56.043±51.277)。结论:半夏泻心汤水溶提取物可能通过降低结肠黏膜IL-5和IL-13的表达而发挥其治疗BALB/c小鼠恶唑酮结肠炎的作用。 Objective:To evaluate the efficacy and mechanism of the water-soluble extracts of Banxiaxiexin decotion in BALB/ c mice with oxazolone-colitis.Methods:On day 1 and day 2,30 BALB/c mice were skin-sensitized with 200μL3% oxazolone and on day6they underwent retention-enema with150μL1%oxazolone,to establish ulcerative colitis model.Then10 of them were treated by intragastric administration with the water-soluble extracts of Banxiaxiexin decotion for 7 days(banxia group);another 10 mice treated with salicylazosulfapyridine(SASP)were taken as the positive-therapy control(SASP group);the other 10 mice were taken as model group.Ten mice not treated with SASP or other drugs were taken as control group.After finishing the therapy,disease activity index(DAI)and histopathological inflammation score were determined,and the expression of interleukin-5(IL-5)mRNA and interleukin-13(IL-13)mRNA in the colonic tissues were detected by fluorescent quantitative reverse transcription polymerase chain reaction(RT-PCR).Results:The DAI and histopathological inflammation score of the model group(1.6±0.3,2.7±0.7)were significantly higher than those of the control group(0.2±0.1,1.7±0.5),and the expression of IL-5 mRNA and IL-13 mRNA in the colonic tissues of the model group(50.8974±23.2601,155.0088±76.6417) were also significantly higher than those of the control group(1.8914±1.2814,4.4826±3.8836).The water-soluble extracts of Banxiaxiexin decotion could significantly lower the DAI(0.3±0.3)and histopathological inflammation score(1.5±0.4).The expression of IL-5 mRNA and IL-13 mRNA in the colonic tissues(7.8222±5.3210,39.9993±22.4196)were also significantly lowered by Banxiaxiexin decotion.Its therapeutic effect was equal to SASP(0.4±0.2,1.3±0.6;21.2973±20.7996,56.0433 ±51.2768).Conclusions:The water-soluble extracts of Banxiaxiexin decotion may exert its therapeutic effect on oxazolone-colitis in BALB/c mice by lowering the expression of IL-5 and IL-13 in the colonic mucosa.
