摘要
提出了毛细管区带电泳法同时测定茶叶中儿茶素、表儿茶素、原儿茶酸、原儿茶醛和槲皮素等5种化合物含量的方法。采用无涂层适应毛细管柱(57cm×75μm,有效长度50 cm)分离,以含10mmol·L^(-1)四硼酸钠、5mmol·L^(-1)β-环糊精、15%(体积分数,下同)乙腈、2%乙二醇pH9.11的缓冲溶液为电泳介质,分离电压为20kV。儿茶素、表儿茶素、原儿茶酸、原儿茶醛和槲皮素线性范围分别为8.1525~217.4,8.1525~217.4,2.174~217.4,1.739~150.0,1.739~150.0mg·L^(-1)。方法用于不同茶叶提取物样品中5种化合物含量的测定,测得平均回收率在94.6%~107%之间,测定值的相对标准偏差(n=5)在0.17%-4.4%之间。
Capillary zone electrophoresis (CZE) was applied to the determination of 5 active components (i. e., protocatechuic aldehyde, protocatechic acid, epicatechin, DL-cateehin and quereitin) in tea. Efficient separation was attained by using the un-coated adaptive quartz capillary column (57 cm×75 μm, effective length 50 cm) and the electrophoretic medium of a buffer mixture, containing 10 mmol ·L-1 Na2B407, 5mmol ·L-1 CD, 15% (φ) acetonitrile and 2% (φ) ethylene glycol (acidity adjusted to pH 9. 11) with separation voltage of 20 kV. I.inearity ranges were found between 8. 152 5-217.4 mg ·L-1 (for protocatechuic aldehyde), 8. 152 5- 217.4 mg·L1 (for protocateehic acid), 2. 174-217. 4 mg· L-1 (for epieatechin), 1. 739- 150. 0 mg· L -1 (for DL-catechin) and 1. 739-150.0 mg ·L 1 (for quercitin). The proposed method was used for determination of the 5 components in tea samples, giving values of recovery and RSD's (n=5) in the ranges of 94. 6%- 107% and 0. 17%-4.4% respectively.
