复方甘草甜素中甘草甜素的含量测定研究

摘要 建立了复方甘草甜素含量HPLC测定方法。采用岛津高效液相色谱仪(SPD-10Avp紫外检测器、LC-10ATvp泵),Anastar工作站,Shimadzu十八烷基硅烷键合硅胶柱(5μm填料,4.6×150mm)。2%冰醋酸-乙腈(65∶35)为流动相,流速为1.0mL/min,检测波长254nm。杂质在上述色谱条件下能很好的分离出,样品在0.042~0.209mg/mL浓度范围内与其峰面积呈良好的线性关系,峰面积A与浓度C的回归方程为:A=1E+06C-11323,R=0.9981;回收率试验的平均回收率为100.10%,RSD为0.67%;精密度试验的RSD为0.10%。本方法快速、简便、结果准确可靠,可作为测定注射用复方甘草甜素的含量的有效方法。 A HPLC method for determination of glycyrrhizin content in compound glycyrrhizin injection was developed. Dikma ODS column was used and column temperature was 40℃, the mobile phase was 2% glacial acetic acid-methyl cyanide(65 : 35), the flow rate was 1.0 mL/min and UV detective wavelength was 254 nm. Results showed: some impurities were obviously separated under above conditions. The calibration curves were linear between 0.042-0.209 mg/mL (r =0.998 0). The average recovery of adding sample was 100.10%, RSD was 0.67%. Precision RSD was 0.10%. The method is quick, simple and sensitive. It would be a valid method for the determination of compound glycyrrhizin for injection and its related substances.
