丹紫康膝冲剂对大鼠膝骨性关节炎软骨Ⅱ型胶原影响的实验研究 被引量:3

Effect of Danzi Kangxi granules on type Ⅱ collagen in articular cartilage among rats with knee osteoarthritis: an experimental study

摘要 目的:观察丹紫康膝冲剂对大鼠膝骨关节炎软骨II型胶原的影响。方法:选取健康Wistar大鼠80只,随机分为丹紫康膝组、西药组、模型组、空白对照组,每组20只。对丹紫康膝组、西药组、模型组大鼠运用改良Hulth造模法进行大鼠膝关节骨性关节炎造模,于造模6周后对各组大鼠进行药物干预。丹紫康膝组予丹紫康膝冲剂灌胃,西药组予硫酸氨基葡萄糖灌胃,模型组、空白对照组均予以等容积蒸馏水灌胃,每天早晚各1次,连续4周。肉眼观察大鼠膝关节形态并按软骨损伤分级法进行分级;光镜下切片观察软骨退变情况,并采用改良Mankin病理骨关节炎评分法评分;免疫组化检测软骨Ⅱ型胶原表达。结果:肉眼观察见空白对照组膝关节软骨表面光滑润泽,模型组造模后膝关节软骨可见局部溃疡性改变、关节边缘增生,西药组和丹紫康膝组可见造模后膝关节生成新生软骨;镜下观察见空白对照组正常,模型组重度退变,丹紫康膝组和西药组轻中度退变;膝关节软骨Ⅱ型胶原含量空白对照组较其余3组高(P<0.05),丹紫康膝组和西药组较模型组高(P<0.05),丹紫康膝组与西药组比较差异无统计学意义(P>0.05)。结论:丹紫康膝冲剂能提高大鼠膝关节软骨Ⅱ型胶原含量,具有防治大鼠早中期膝骨性关节炎软骨退变的作用。 Objective : To observe the effect of Danzi Kangxi granules on type II collagen in articular cartilage among rats with knee osteoarthritis. Methods : Eighty healthy Wistar rats were randomly and equally divided into Danzi Kangxi group, Western medicine group, model group, and blank control group. A rat model of knee osteoarthrifis was established by improved Huhh method in the Danzi Kangxi group, Western medicine group, and model group, and each group received medication at 6 weeks after the model was established. The Danzi Kangxi group received Danzi Kangxi granules by gavage, the Western medicine group received glucosamine sulfate by gavage, and the model group and blank control group received an equal volume of distilled water by gavage. Medication was given once in the morn- ing and once in the evening for 4 consecutive weeks. The morphology of knee joint was observed with the naked eye and was subjected to cartilage injury grading. Cartilage degeneration was evaluated by observing sections under a light microscope, and the severity of oste- oarthritis was evaluated using the modified Mankin score. The expression of type II collagen in articular cartilage was measured by im- munohistochemistry. Results:The naked -eye observation showed that the surface of knee articular cartilage was smooth and moist in the blank control group ,the knee articular cartilage had local ulcerative changes and articular edge hyperplasia in the model group, and the knee joint had newly formed cartilage in the Western medicine group and Danzi Kangxi group. The microscopic observation showed that the blank control group had normal articular cartilage, the model group had severe cartilage degeneration, and the Danzi Kangxi group and Western medicine group had moderate cartilage degeneration. The blank control group had a significantly higher content of type II collagen in knee articular cartilage than the other three groups (P 〈 0. 05 ) ;the Danzi Kangxi group and Western medicine group had a significantly higher content of type II collagen in cartilage than the model group (P 〈 0. 05 ) ;there was no significant difference in content of type II collagen between the Danzi Kangxi group and Western medicine group(P 〉 0. 05 ). Conclusion: Danzi Kangxi granules can increase the content of type II collagen in knee articular cartilage among rats and are capable of preventing and treating the cartilage degeneration in rats with middle - stage knee osteoarthritis.
