摘要
目的观察长春新碱对人成骨肉瘤MG63细胞增殖的抑制及凋亡促进作用,探讨其作用的分子机制。方法以0、5、10、20、50、100μg·L-1不同浓度长春新碱处理的MG63细胞,分别经CCK-8及Hoechest 33342检测细胞增殖和细胞凋亡水平,用实时定量RT-PCR分析mRNA表达水平。结果长春新碱可抑制MG63细胞增殖;长春新碱可以促进细胞凋亡;长春新碱可以下调c-myc及上调caspase9基因表达(P<0.05)。结论长春新碱从较低浓度起即有抑制细胞增殖及促进细胞凋亡作用,其作用机制是通过下调c-myc及上调caspase9的基因表达实现的。
Objective To explore the proliferation inhibiting and apoptosis inducing effect of vincristine on human osteosarcoma MG63 cells and clarify the molecular mechanisms. Methods Cell number counting were determined with CCK8 and cell apoptosis were determined with Hoechest 33342 ,respectively in MG63 cells which explored on different concerntrations of vincristine (0,5,10,20,50,100 μg · L 1 ). Real time quantitative RT-PCR was used to analyze gene expression. Results 10vg. L 1 vincristine could significantly inhibit the proliferation of MG63 cells (P〈0.05) ,the apoptotic cells increased with the increasing of concentration of vincristine; vincristine decreased the expression of c-myc gene (P〈0.05) and increased the expression of caspase9 gene (P〈0.05). Discussion: Vincristine can inhibit the proliferation and promote the apoptosis of MG63 cells with low concentration,its mechanism is to down-regulate the c-myc and up-regulate the caspaseg.
