黄山风景区截获线虫分离物鉴定 被引量:1

摘要 黄山松(Pinus taiwanensis Hayata)是分布于我国东南沿海的一种重要松属树种,也是构成黄山风景区重要生态景观树种.为防控松材线虫病,途经黄山的木质包装材料都必须进行分离物检查鉴定.作者运用分子鉴定、形态鉴定等手段,对黄山风景区检疫检查站截获的一株线虫分离物进行了鉴定.采用松材线虫特异引物,对检疫检查站截获的虫株进行分子检测,样品中不合松材线虫.通过对分离物的形态测量与比对,鉴定为程氏伞滑刃线虫(Bursaphelenchus chengi).雌、雄虫线形,雌虫长(769.28土22.72)μm,雄虫长(757.60±28.40) μm.唇区高,头部和身体界线明显.口针细,长15~ 16 μm,基部没有膨大.中食道球发达,近圆球形,占体宽的2/3以上.雌成虫阴门位于虫体3/4处,有较小的阴门盖.雌成虫尾部为圆锥形,尾端钝圆,无明显的尾尖突.雄成虫交合刺大,末端三分之一处强烈向腹面弯曲,基顶高,喙突明显,指状,远端有盘状突起,交合伞铲状. Pinus taiwanensis is an important tree species of Pinus,which distribute in the southeast coastal area in China,and also this tree specie forms the ecological landscape of Huangshan scenic area.Thus the wood packaging material which delivered through the rigion must be isolated and inspected.By means of techniques such as molecular identification,morphological identification technique,has identified nematode isolates captured by the Quarantine Inspection Station of Huangshan scenic area.After the molecular testing on the nematode isolate by using specific primers of Bursaphlenchus xylophilus,we found that the sample did not contain B.xylophilus.The morphological identification showed that the nematode isolate at Huangshan mountain scenic area was B.chengi.Both female and male are line-shape,with the length of female (769.28 ± 22.72) μm,and the length of male (757.60 ± 28.40) μm.Lip region is high,rounded.The stylet length is about 15-16 μm long,and basal knobs absent.Median bulb is elongate-oval,which accounts for more than two-thirds of the body width.The vulva of adult female is at 3/4 area of the whole body,covered by a relatively small vulva cover.The end of its tail is obtuse,without obvious tail tips.Male adult has a big spiculum and the 1/3 part of its tail is bending to the stomach.Its base is tall with obvious coracoid,which is finger-shape.Apex of bursa is concave.
