摘要
目的:建立氯化铁外伤性癫痫模型后,结合不同检测方法和不同时间点来动态判断自噬活性。方法:SD大鼠根据实验目的随机分为6组,分别用LC3Ⅱ/LC3Ⅰ,自噬体计数,Bcl-1不同自噬检测方法检测自噬活性,收集实验数据采用单因素方差分析进行统计学分析。结果:通过统计分析得出1 h内可见LC3-II/I比值为(3.02±0.001),与对照组相比,P<0.05;随时间推移,峰值位于1d,其中LC3-II/I比值并未进行性升高,反而有下降趋势,P<0.05;自噬体计数与对照组相比,与前一时间点相比,自噬体计数进行性增加,P<0.05;1 h组Bcl-1达到峰值,随后逐渐减少,P<0.05;癫痫模型建立后Bcl-1表达与对照组相比,P<0.05。结论:LC3Ⅱ/LC3Ⅰ,自噬体计数,Bcl-1均可用来判断自噬活性,评价自噬动态活性应结合两种或两种以上方法进行检测。
Objective: To establish the animal model of traumatic epilepsy ferrous chloride, a combination of dif ferent detection methods and different time points to dynamically determine autophagic activity. Method according to the experimental SD rats were divided into 6 groups, respectively by LC3 II / LC3 I, autophagy body counts, Bcl-1 different autophagy detection method to detect autophagic activity, collect experimental data to show, and the use of the single factor variance analysis of statistical analysis. Results through statistical analysis of 1 hours of the LC3-II / I ratio of 3.02 + / - 0.001, pmpared with the control group, P 〈 0.05, with the passage of time, peaking at 1 day, in which LC3-II / I ratio were increased, but decreased, P 〈 0.05; the autophagosome count and PI count is compared with the control group, with the previous time points compared, autophagosome counts were increased, P 〈 0.05; group lh Bcll reaches a peak, then gradually decreased, P 〈 0.05 , epilepsy model established Bcll expression compared with control group, P 〈 0.05. Conclusion LC3 II / LC3 I, autophagy body counts, Beclin Bcl 1 can be used to determine the autophagic activity evaluation of autophagy, dynamic activity should be combined with two or more than two kinds of methods for testing.
