低氧对大鼠骨髓间充质干细胞粘附能力的影响 被引量:2

摘要 目的探讨低氧对大鼠骨髓间充质干细胞粘附能力的影响。方法采用SD大鼠20只,年龄6~8周、雌雄不限、体质量100~120g,获取骨髓间充质干细胞。流式细胞鉴定实验细胞CD54、CD29、CD90、CIM5表达。根据细胞培养及冲涮实验时的细胞氧环境,实验分组为4组:A组(20%O2培养20%O2冲涮组);B组(1%O2培养1%O2冲涮组);C组(20%O2培养1%O2冲涮组)和D组(1%O2培养20%O:粘附冲涮组),比较不同组下剪切力冲涮细胞的残留率。研究不同氧浓度变化对骨髓间充质干细胞表达缺氧诱导因子-1α(hypoxia inducible factor,HIF-1α)和血管细胞粘附分子(vascularcell adhesion molecule,VCAM-1)的影响。结果骨髓间充质干细胞在1%O2环境下(B、C组)粘附能力增强,在20%O2环境下(A、D组)粘附能力减弱(P〈0.05);HIF-1α表达水平在1%O2环境下(B、C组)增强(P〈0.05)。结论骨髓间充质干细胞随着粘附时问越长,细胞粘附能力也逐渐增强,经过低氧处理细胞的粘附力提高时间快于常氧环境。
