血管性痴呆小鼠海马组织GSK3β、p-GSK3β蛋白的动态表达及意义 被引量:4

摘要 目的观察血管性痴呆(vascular dementia,VD)小鼠海马组织糖原合成酶激酶3β(glycogen synthase kinase-3β,GSK3β)和p-GSK3βSer9蛋白表达的动态变化及意义。方法采用反复3次间断结扎双侧颈总动脉的方法建立VD小鼠模型,并设假手术组,术后2周、4周、6周利用水迷宫实验和跳台实验观察小鼠行为学改变,采用HE染色观察海马组织病理学变化,Western-blot方法检测小鼠海马组织GSK3β、p-GSK3βSer9蛋白的表达变化。采用析因设计的方差分析进行统计分析。结果水迷宫试验中,小鼠学习阶段游完全程时间的处理×时间交互效应没有统计学意义(P>0.05),但记忆阶段2者交互效应有统计学意义(P<0.05)。跳台实验中,小鼠反应时间和潜伏时间的处理×时间交互效应无统计学意义(均P>0.05)。同时,HE染色显示模型组小鼠海马组织神经元减少,4周、6周时可见细胞核固缩。各组小鼠GSK3β总蛋白表达的主效应和交互效应均无统计学意义(P>0.05);各组小鼠p-GSK3β蛋白表达的,处理主效应、时间主效应和2者交互效应均有统计学意义(均P<0.001)。结论反复脑缺血可造成小鼠持续的学习记忆功能障碍,海马组织受损严重,p-GSK3β蛋白的动态变化参与血管性痴呆的发生过程。
