摘要
目的探讨脂肪来源干细胞(ADSCs)对光老化成纤维细胞的治疗作用。方法分离培养小鼠ADSCs,并进行表面标志物鉴定;ADSCs与成纤维细胞(FBs)共培养48h后,单次UVB120mJ/cm2照射后30min检测细胞内活性氧(ROS)生成量;采用UVB120mJ/cm2反复照射4次,照射间隔时间12h,诱导产生细胞光老化模型,继续培养48h后检测细胞增殖能力、合成弹性蛋白原等变化。结果体外培养ADSCs呈成纤维细胞样,活力良好,表面标志物CD29、CD90.2和sca-1阳性,CD34和CD45阴性;ADSCs可减轻单次UVB照射引起的氧化应激损伤,保护细胞免受细胞周期阻滞的影响,同时可促进光老化FBs合成弹性蛋白原。结论ADSCs对光老化FBs具有保护作用,可作为对抗光老化的新手段。
Objective To investigate the therapeutic effect of adipose-derived stem cells (ADSCs) on photo-aging fibroblasts (FBs). Methods ADSCs were isolated and cultivated in vitro, identified by surface markers. After co-culture with transwell chambers for 48 hours, FBs were irradiated by a series of 4 UVB expo- sure at the dose of 120 mJ/cm2. The generation of reactive oxygen species (ROS) was detected immediately af- ter one single exposure of UVB at 120 mJ/cm2. The proliferation capacity and secretion of tropoelastin were e- valuated 48 hours after the last exposure with co-culture of ADSCs. Results ADSCs were fibroblast-like cells in culture, maintaining satisfactory cell viability. ADSCs were positive for CD29, CD90.2, Sca-1, and negative for CD34 and CD45. The generation of ROS was reduced by ADSCs co-culture. The photoaging FBs showed cell cycle arrest while ADSCs counteracted this effect. Regarding secreting function, ADSCs rescued the pho- toaging FBs by enhancing tropoelastin synthesis. Conclusion ADSCs were here demonstrated protective role on photoaging FBs, and could serve as a new potential therapy in anti-photoaging skin.
