摘要
目的观察碱性成纤维细胞生长因子(bFGF)在多孔高密度聚乙烯(Medpor)-软骨复合支架血管化中的应用效果,探讨其使用意义。方法构建兔耳软骨与Medpor复合支架模型,植于兔背部皮下,将实验动物分成2组:耳软骨与Medpor材料直接连接组(A组);耳软骨与Medpor材料直接连接后,bFGF处理组(术中及术后2d,每日局部注射bFGF100ng,B组);置入后第3、7、14、28天后取出复合支架,行大体和组织学观察,免疫组织化学法(SABC法),检测Medpor片边缘及内部CD34表达,并行微血管密度(MVD)计数。结果B组术后早期Medpor材料周边纤维组织增生及血管化较A组明显,但后期血管化程度与A组无明显差异。结论术后早期应用bFGF对复合支架的纤维血管化过程中起到明显的促进作用,加强Medpor与软骨片的连接,增强复合支架的稳定性及抗感染力。
Objective To observe the application effect of basic fibroblast growth factor (bFGF) in the medpor-cartilage scaffold vascularization, and explore its significance. Methods Build rabbit ear cartilage with Medpor bracket model, implanted in rabbits subcutaneously, and the animals were divided into two groups : the ear cartilage and Medpor material directly connected group ( Group A ) ; the ear cartilage directly connected with medpor, and the scaffold was pretreated with bFGF( 100 ng/d×3 d, Group B). At the 3, 7, 14, 28 clays after implantation, remove the composite scaffold for gross and histological observation, immuno- histochemical method (SABC method ) detection of CD34 expression in composite scaffolds and parallel mi- crovessel density (MVD) count. Results Proliferation of fibrous tissue and vascularization in the early postop- erative Group B than Group A was significantly, but no significant difference in the late vascularization with group A. Conclusion Early postoperative application of bFGF play a significant role in promoting for compos- ite scaffolds fibrovascular process, and strengthen the connection between the Medpor material and cartilage piece, enhance the stability of the composite scaffold and anti-infectious.
