灯盏花素对大鼠胰腺缺血再灌注损伤的保护作用的实验研究

摘要 目的:探讨灯盏花素对大鼠胰腺缺血再灌注(ischemia-reperfusion,I/R)损伤有无保护作用及其可能机制。方法:制作大鼠胰腺I/R模型,45只SD大鼠随机分为假手术组(A组)、I/R组(B组)、灯盏花素组(C组),各组15只。采用ELISA和免疫组化方法分别检测各组大鼠胰腺缺血30 min再灌注0、6、12 h时血清中肿瘤坏死因子α(tumor necrosis factorα,TNF-α)、白细胞介素1β(interleukim 1β,IL-1β)的含量和胰腺组织中Survivin蛋白表达的变化;同时观察胰腺组织病理学变化。结果:B组胰腺组织病理学改变较其他组明显;在再灌注0、6、12 h时B、C组血清中TNF-α、IL-1β含量均较A组显著升高(P<0.05),但C组增高水平明显低于B组(P<0.05);C组在再灌注6、12 h时Survivin蛋白表达明显高于A、B组(P<0.05),A、B组比较差异无显著性(P>0.05)。结论:灯盏花素能降低大鼠胰腺I/R损伤时血清TNF-α、IL-1β水平和上调凋亡抑制基因Survivin蛋白表达,对大鼠胰腺I/R损伤有明显保护作用。 Objective: To study the effect of Breviscapin during isehemia-reperfusion injury of rat pancreas and assess the possi- ble mechanism, nothods: 45 SD rats were randomly divided into pseudoperation group (group A), ischima-reperfusion group (group B) and breviscapin group (group C). During the pancreas ischemia 30 minutes and reperfusion 0, 6, 12 hours, the level of tumor necrosis factor α (TNF-α), interleukin 1 β (IL-1β) in the serum were determined by enzyme linked immunosorbent assay (ELISA). Expression of Survivin in pancreas celles were studied by immunohistochemical methods. Pancreas histopathology was ob- served respectively. Results: There were evident pathological changes in group B. The expression level of TNF-α, IL-1 β in groups B, C was obviously higher than those in group A (P 〈 0. 05 ). But the level of TNF-α, IL-1 β in groups C was obviously lower than those in group B (P 〈0. 05). The expression level of Survivin in group C was higher than those in groups A, B at 6 h and 12 h of reperfusion (P 〈 0.05). But there was no significant difference between group A and group B. Conclusions: Breviscapin can re- duce the expression level of TNF-α and IL-1β during the pancreas ischemia and reperfusion, and can enhance the expression level of Survivin. It has protective effects on ischemia-reperfusion injury of rat pancreas.
