摘要
目的:探讨降糖通络方对2型糖尿病大鼠坐骨神经功能、血管活性因子及糖脂代谢紊乱的影响。方法:采用高脂乳剂灌胃联合尾静脉注射链脲佐菌素(STZ)制备大鼠2型糖尿病模型,随机分为模型组、阳性药组(二甲双胍0.26 g/kg+弥可保0.135 mg/kg)、降糖通络方组,并以正常组做对照。给药6周后测定各组大鼠空腹血糖(FPG)、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL)、高密度脂蛋白(HDL)、内皮素(ET)及一氧化氮(NO)水平以及大鼠坐骨神经传导速度和热刺激敏感性。结果:与模型组比较,阳性药及降糖通络方组大鼠FPG、血清TC、TG、LDL和ET水平显著降低,HDL和NO水平、坐骨神经传导速度及热刺激甩尾潜伏期显著升高(P<0.05)。结论:降糖通络方对2型糖尿病大鼠外周神经病变具有保护作用,其可能与稳定糖脂代谢紊乱和平衡血管活性因子有关。
Objective : To investigate the effects of Jiangtang Tongluo Formula on the function of sciatic nerve, levels of vascular active factors and the disorder of lipid in type 2 diabetic rats. Method : Type 2 diabetic rat model was established by high - fat diet with tail vein injection of streptozotocin (STZ). Model rats were randomly divided into model group, positive medicine group (metformin 0.26 g/kg + methylcobalamin 0. 135 mg/kg) , Jiangtang Tongluo Formula group and control group. Six weeks later, the fasting plasma glucose ( FPG), total cholesterol ( TC ), triglyceride ( TG), low density lipoprotein( LDL), high density lipoprotein(HDL) , endothelin(ET) and nitric oxide(NO) levels, as well as the sciatic nerve conduction velocity and thermal stimulus sensitivity were measured. Result : Compared with model group, the levels of FPG, TC, TG, LDL and ET were significantly reduced while the levels of HDL and NO, sciatic nerve conduc- tion velocity and the tail - flick latency for heat - stimulation were significantly higher(P 〈 0.05 ). Conclusion : Jiangtang Tongluo Formula has a protective effect on peripheral neuropathy in type 2 diabetic rats, which may be related with its effect on stability of lipid disorders and balance of vascular active factors.
