摘要
目的 探讨Exendin-4在高脂状态下是否具有抗脂毒性及其相关的机制是否涉及葡萄糖激酶(GK)和葡萄糖转运蛋白2 (GLUT2). 方法 应用不同浓度Exendin-4(5、10及50 nmol/L)孵育βTc6细胞不同时限(6、12及24 h),再予1mmol/L FFA作用24h后,检测细胞内GK和GLUT2表达情况.结果 Exendin-4干预6h,各组细胞内GK、GLUT2表达未见明显改变;当干预延长至12~24 h,随干预浓度增高,细胞内GK、GLUT2表达逐步升高. 结论 适宜浓度的Exendin-4干预一定的时限可上调脂毒性状态下胰岛β细胞GK和GLUT2的表达.
Objective To investigate the anti-lipotoxicity effect of Exendin-4 on lipotoxic state and whether its mechanism is related with glucokinase (GK) and glucose transporter 2 (GLUT2). Methods βTc6 cells were incubated with Exendin-4 (5,10 and 50 nmol/L) for different duration (6,12 and 24 h) prior to intervention with 1 mmol/L free fatty acids (FFA) for 24 hours,then OK and GLUT2 expressiqn was examined. Results When treating with Exendin-4 for 6 hours, the expression of GK and GLUT2 in βTc6 cells was not up-regulated in each group. After treating with Exendin-4 for 12 - 24 hours, the expression of OK and GLUT2 was increased gradually along with increasing the concentration of Exendin-4. Conclusion Treating with Exendin-4 in optimal concentration and duration, the expression of GK and GLUT2 can be up-regulated in lipotoxic state.
