摘要
目的通过建立大鼠慢性酒精中毒自由饮模型,观察和探讨慢性酒精中毒性视网膜及视神经病理改变。方法 36只Wistar雄性大鼠随机分成对照组、实验组,各18只。其中对照组饮用普通水;实验组酒精浓度从6、9、12%各5d递增至20%后以该浓度维持饲养。于第2月、3月、4月末行视网膜及视神经病理检查。结果实验组大鼠在3月及4月末视网膜单位长度内神经节细胞减少,与对照组相比差异具有显著性(P<0.01)。3月及4月末实验组及对照组视神经半薄切片甲苯胺蓝染色结果示实验组视神经单位面积内神经纤维减少,与正常组相比差异具有显著性(P<0.01)。结论大鼠慢性酒精中毒自由饮模型可用于模拟人类慢性酒精中毒性视网膜及视神经病变。
Objective Using a free-drink model of chronic alcoholism in rats to study the pathological changes of chronic alcoholism on the retina and optic nerve.Methods Thirty-six wistar male rats were randomly divided into two groups: control group and ethanol group,each group enrolled eighteen rats.The control group fed with water and general diet.The ethanol group fed with general diet and water which contained alcohol concentration of 6% 、9% 、12%(V/V),each concentration lasted for 5 days,then the alcohol concentration changed for 20%(V/V).The pathological examination were taken at the end of 2 months、3 months and 4 months.Results The retinal ganglion cells(RGCs) of the rats in ethanol group were significantly less than that of the control group(P 0.01) and the number of axon per unit area in ethanol group were significantly different with control group(P 0.01) at the end of 3 and 4 months.Conclusion The free-drink model of chronic alcoholism can be used for the research of the change of human retinal and optic nerve in chronic alcoholism.
