摘要 为了探索坦布苏病毒E蛋白的抗原结构,以鹅源坦布苏病毒SHYG株E蛋白基因组序列为基础,用DNAStar、Antheprot等软件对E蛋白的二级结构、亲水性、表面可能性、抗原性指数、跨膜结构及等电点等进行预测,用I-TASSER分析E蛋白三级结构。结果表明坦布苏病毒E蛋白等电点为7.15;综合分析显示37~39、80~86、124~126、133~134、233~237、276、315~318、398~399位是可能的B细胞抗原表位,其中80~86、233~237、315~318、398~399位形成抗原表位可能性更高。抗原表位的预测为坦布苏病毒E蛋白的抗原表位的鉴定提供理论依据。 To investigate the antigenic characteristics of E protein of Tembusu virus,E gene sequence of a goose-origin Tembusu virus strain SHYG was selected.The secondary structure,hydrophilicity plot,surface probability plot,antigenic index,transmembrane region,and isoelectric point were predicted by DNAStar and Antheprot.The tertiary structure of E protein were predicted by the on-line platform I-TASSER and viewed by RasMol.The analysis results showed that the isoelectric point of E protein was 7.15,and the amino acid residues 37 to 39,80 to 86, 124 to 126,133 to 134,233 to 237,276,315 to 318,and 398 to 399 were deduced as B cell epitopes for E protein,and 80 to 86,233 to 237,315 to 318,398 to 399 were predominant epitopes. This study provided a theoretical foundation for identification of antigenic epitopes.
