摘要
目的:建立一种快速溶剂萃取(ASE)-高效液相色谱检测辣椒中辣椒素和二氢辣椒素的新方法。方法:以甲醇-四氢呋喃(1:1)为萃取剂,考察了萃取温度、萃取时间、萃取循环次数3个因素对ASE萃取效果的影响,随后用高效液相色谱进行检测,并将该方法与国标方法GB/T 21266—2007和药典方法《中国药典》2010版(一部)进行了对照试验。结果:最佳萃取工艺:萃取温度为60℃、萃取时间为20 min、萃取循环次数2次。在此条件下用高效液相色谱法检测了4种辣椒样品中辣椒素和二氢辣椒素的含量,检测结果同国标方法较一致,且优于《中国药典》2010版(一部)。结论:建立的快速溶剂萃取(ASE)-高效液相色谱检测方法具有高效、自动化、无毒、快速的特点,对辣椒中辣椒素和二氢辣椒素的含量检测取得了良好效果,是一种具有实用价值的分析方法。
Objective: A method based on accelerated solvent extraction (ASE) HPLC was developed for the determination of capsaicin and dihydrocapsaicin in capsicum annuum L. Methods: Using the methanol tetrahydrofuran(l:l) as solvents, the extractions efficiency of ASE were studied, inspecting the temperature of extraction, the time of extraction, the cycle times of extraction. Then the capsaicin and dihydrocapsaicin were determined by HPLC. In addition, the check experiments were analyzed from ASE to GB/T 21266-- 2007 and Chinese pharmacopoeia 2010 edition. Results: The optimum extracting technology was as follows: the temperature of extraction was 60 %, the time of extraction was 20 min; the cycle times of extraction was 2 times. The contents of capsaicin and dihydrocapsaicin in four kinds of capsicum annuum determined by HPLC were close to GB/T 21266--2007 and preceded Chinese pharmacopoeia 2010 edition. Conclusion: The characteristics of ASE-HPLC are high efficient, automatic, non-toxic and quick. So this method has practical value and can use for analysis.
