摘要
目的探讨p38MAPK抑制剂SB202190体外对细粒棘球蚴原头节生长的作用。方法将体外培养的细粒棘球蚴原头节分别加入12.5、25、50、100μmol/L的SB202190中体外孵育,在光镜下观察原头节的形态变化,同时通过伊红染色显示原头节活力。实验重复3次。结果孵育14d后,正常和DMSO对照组原头节的活力几乎没有改变。50μmol/L和100μmol/L SB202190作用1d后,细粒棘球蚴原头节的活力开始下降;作用14d后,100μmol/L SB202190组无存活的头节,50μmol/L SB202190组的头节活力仅为13.8%。12.5μmol/L和25μmol/L SB202190对原头节的活力也有影响,但不及高浓度组显著。结论 p38MAPK抑制剂SB202190在体外有抗细粒棘球蚴原头节的作用。
Objective The aim of this study was to investigate the in vitro efficacy of the p38MAPK inhibitor SB202190 against Echinococcus granulosus protoscoleces.Methods Protoscoleces of E.granulosus were incubated in vitro with SB202190at concentrations of 12.5,25,50,and 100μmol/L,and the viability of protoscoleces was assessed on a daily basis by microscopic observation of movements,flame cell activity,and 0.1% eosin staining.Each experiment was repeated three times.Results Control E.granulosus protoscoleces incubated in RPMI 1640medium or in RPMI 1640 medium+DMSO remained viable after 14days of incubation.From day 1,the viability of protoscoleces started to decline due to the 50μmol/L or 100μmol/L of SB202190.After 14days,viable protoscoleces were no longer observed in cultures treated with 100μmol/L of SB202190but survival of 13.8%of protoscoleces was observed when they were treated with 50μmol/L of SB202190.Although 12.5μmol/L and 25μmol/L of SB202190also had an effect on protoscoleces,the effect was not as pronounced as that of 100μmol/L of SB202190.Conclusion Results indicated that SB202190has satisfactory action against E.granulosus protoscoleces in vitro.
