摘要
以苦马豆子叶为外植体,培养在MS培养基上诱导愈伤组织,在初步筛选激素组合的基础上,运用二次回归正交设计方法,对愈伤组织组织培养体系建立中的关键因素进行优化.结果显示,2,4-D与Kt的不同浓度组合方式有显著差异,根据立回归方程的结果,两种激素的最优组合结果为1.79 mg·L-1的2,4-D和1.15mg·L-1的Kt,理论最大鲜重为0.9006g.经实验,培养36d后,每块愈伤组织鲜重可达0.9752g,与理论值无显著差异.结果表明二次回归正交设计方法可成功用于优化植物愈伤组织诱导诱导条件.
Using cotyldon of Swainsona salsula as explant for callus induction on MS medium,the key inducing factors were opimized by quadratic regressive orthogonal design,based on primarily screened homone combintions.The results showed that significant differenc existed in the combinations of 2,4-D and Kt,and their optimal concentrations were elationship of the concentration of 2,4-D and Kt and their combination were 1.79 mg · L-1 and 1.15mg · L-1,respectivley.Callus cultured on MS medium with this combination reached 0.9752 g with fresh weight,after 36 days of culture.The results indicated that the quadratic regressive orthogonal design was applied successfully for optimizing conditions for callus induction.
