Expressions and Significance of Thymidine Kinase 1 and Prognostic Factors in Primary Breast Cancer Tissues

摘要 目的探讨TK1与乳腺癌预后相关因子(Ki67、ER、PR、C-erbB-2、P53)的表达关系及意义。方法选取广州医学院第二附属医院2005年6月至2009年6月间手术切除的乳腺癌患者组织标本65例,均经病理证实。用免疫组化分别测量TK1与Ki67、ER、PR、C-erbB-2、P53表达情况。结果在65例乳腺癌组织中,TK1阳性表达率为81.5%(53/65),Ki67阳性表达率为61.5%(41/65),ER、PR、C-erbB-2和P53阳性表达率分别是:64.6%(42/65),67.6%(44/65),60.0%(39/65)和46.2%(30/65)。TK1阴性和阳性表达在不同年龄、TNM分期、组织学分级、腋窝淋巴结转移状况和肿块大小中无显著性差异(P>0.05)。经Spearman相关分析发现TK1与Ki67成正相关,相关系数r=0.520,P<0.05。然而,TK1与ER、PR、C-erbB-2、P53无相关性(相关系数r=0.054,P>0.05)。结论 TK1能准确反应细胞增殖,有助于判断乳腺癌预后。 Objective To investigate the expressions and significance of Thymidine Kinase 1(TK1) and prognostic factors(Ki67,ER,PR,C-erbB-2,P53) in primary breast cancer tissues.Methods 65 samples of breast cancer tissues were selected from the Second Affiliated Hospital of Guangzhou Medical University from June 2005 to June 2009 randomly,and they were resected by surgical operation and confirmed as breast carcinoma by pathology.Immunohistochemical staining assay was used to detect the expression of TK1,Ki67,ER,PR,C-erbB-2,and P53.Results TK1 was highly expressed in 53 out of 65 breast carcinoma tissues(81.5%).The positive rate of Ki67,ER,PR,C-erbB-2,and P53 were 61.5%,64.6%,67.6%,60.0%,46.2%.The expression of TK1 was not statistically significant in some aspects: age,TNM stage,histology grade,lymph nodes metastasis of fossa axillaris,and tumor size.Moreover,Spearman correlation assay was applied to analyze that the positive expression of TK1 was significantly and positively correlated with Ki67(r = 0.520,P 0.05),just not ER,PR,C-erbB-2,and P53(P 0.05).Conclusions The positive expression of TK1 combined with Ki67 may be a dangerous sign contacted with tumor metastasis and recurrence,and thus helps for the diagnosis and prognosis of breast cancer.
