摘要
[目的]对脂肪酶产生菌黑曲霉ZW-1进行复合诱变育种研究。[方法]以脂肪酶产生菌黑曲霉ZW-1为出发菌株,通过紫外线、微波及硫酸二乙酯进行复合诱变得到变异株,对筛选出的变异株产生的脂肪酶活力进行测定。[结果]试验选育到1株脂肪酶产量较高的变异株8632;在适宜条件下,该菌株产生的脂肪酶活力达104.62 IU/ml,比出发菌株提高了125.86%;遗传稳定性试验表明该突变株具有良好的传代稳定性。[结论]该研究为满足脂肪酶在工业生产中的需求提供了科技支持。
[Objective] The aim was to study the compound mutation breeding of lipase-producing strain Aspergillus niger ZW-1.[Method] Using the lipase-producing strain A.niger ZW-1 as starting strain,mutants were obtained and selected by the compound mutation of ultraviolet,microwave and diethyl sulfate (DES),and the lipase activity of mutants were determined.[Result] A mutant strain 8632 was bred with high yield of lipase; the activity of lipase produced by that strain reached 104.62 IU/ml under suitable conditions,and it was about 125.86% higher than that of the starting strain ; the genetic stability experiment showed that the mutant 8632 had good genetic stability after 5 times of subculture.[Conclusion] The study provided scientific support to meet the needs of lipase in industrial production.
