酵母甘露聚糖的研究(Ⅰ)——纯化与鉴定 被引量:6

摘要 用稀碱从废啤洒酵母里提取一粗多糖,经费林试剂络合沉淀,多次醇析和脱蛋白得一级份 SPI。通过 P.C.和 G.C.检查为一甘露聚糖,醋酸纤维素膜电泳、超离心分析和 Sepharose 4B 柱层析证明 SP1是均一的,分子量约为22万。氨基酸分析和β一消除指出 SP1含17种氨基酸,糖与蛋白质是通过糖的还原端与 Ser 或 Thr 形成 O—糖苷键连接的。 The crude polysaccharides were extracted from beer yeast mud with a dilute base and fractionated by Fehling's solution and ethanol.The free proteins were removed and the frac- tion,SP1,was obtained.P.C.and G.C.analysis indicated that SP1 is a kind of mannan. Cellulose acetate film eleetrophoresis,sedimentation analysis and chromatography on Sepharose 4B showed that SP1 is homogeneous and moleculasr weight is approximately 220,000.Amino acid analysis and β-elimation explained that SP1 contains seventeen kinds of amino acid and the polysaccharides combine proteins by forming O-glycosidic bonds through Ser or Thr.
