多重荧光RT-PCR在手足口病病原检测中的应用分析 被引量:3

摘要 目的多重荧光RT-PCR技术能有效地进行手足口病病原检测。方法应用多重荧光RT-PCR技术,对疑似临床重症、轻症手足口病病例进行病原检测,包括肠道通用病毒、肠道病毒71型和柯萨奇病毒A16型进行检测。结果通过对584个病例,851件样本的检测。其中24个病例EV阳性(未分型),占4.38%;285个病例EV71阳性,占52.01%;120个病例CoxA16阳性,占21.90%;其余核酸检测结果为阴性。其检测结果与普通荧光RT-PCR差异无统计学意义(P>0.05)。结论多重荧光RT-PCR可应用于手足口病病原监测及应急检测工作,尤其是在大样本量的情况下,不仅在一定程度上可以减少耗材的支出,而且可以有效减少人力支出。 Objective To detect the pathogens of hand, foot and mouth disease (HFMD) by multiple fluorescence RT - PCR. Methods The specimens from suspected severe and mild HFMD cases were detected by multiple fluo- rescence RT - PCR for enterovirus 71( EV71 ), Coxsackievirus A16 (CVA16) and the general enterovirus excluding EV71 and CVA16 (EV). Results In 851 specimens from 548 patients, twenty - four cases were found EV positive (untyped), accounting for 4.38%; Two hundred and eighty- five cases were EV71 positive, accounting for 52.01%; One hundred and twenty cases were CoxA16 positive, accounting for 21.90%; the rest were negative. There was no significant difference of the positive rate between the multiple fluorescence RT - PCR and single fluo- rescence RT - PCR (P 〉 0. 05 ). Conclusion The multiple fluorescence RT - PCR can be used for pathogenic di- agnosis of HFMD in normal monitoring and emergency detection, especially in detection of a large samples, which can reduce the costs of supplies and labor.
