灰色链霉菌产链霉素发酵培养基的优化 被引量:1

摘要 通过单因素试验考察了发酵培养基中不同的碳、氮源对灰色链霉菌产链霉素的影响;在此基础上通过正交试验对发酵培养基中4种主要组分,即玉米淀粉、黄豆饼粉、硫酸铵、磷酸氢二钾的用量配比进行优化筛选。结果表明,发酵培养基中碳、氮源及主要无机盐的用量对发酵液中链霉素的产量均有显著影响。4种因素中对产量影响最大的是玉米淀粉浓度,其次是K2HPO4浓度,再次是氮源即黄豆饼粉和硫酸铵的浓度。优化得到的最佳发酵培养基组成为:黄豆饼粉5%、玉米淀粉4%、硫酸铵0.6%、K2HPO41%、MgSO4·7H2O 0.075%、NaCl 1.0%,pH7.8。 The effect of the carbon and nitrogen sources in fermentation medium on streptomycin production of streptomyces griseus was investigated. On this basis, orthogonal design (L9 (34) ) was adopted to optimize the fer- mentation medium of S. griseus. The four main factors were corn starch, soybean cake meal, ammonium sulfate and K2HPO4 concentration. The results suggest that the concentration of carbon source, nitrogen source and main inorganic salt has a significant effect on streptomycin productivity of broth. The sequence of effect was corn starch 〉 K2 HPO4 〉 soybean powder and ammonium sulfate. The composition of an optimized medium for the production of streptomycin was soybean cake meal 5% , corn starch 4% , ammonium sulfate 0. 6% , K2HPO4 1% ,MgSO4 · 7H2O 0. 075% ,NaCl 1.0% ,pH 7.8.
