E3 protein: a key anti-interferon immuneevasion factor of vaccinia virus

摘要 痘苗病毒能够对抗宿主的抗干扰素作用是其致病机理中的一个重要方面。E3蛋白是由痘苗病毒编码的一类重要的抗干扰素免疫逃避因子,它通过其羧基末端的dsRNA结合域与dsRNA作用后抑制干扰素产生系统中的PRK、2′,5′-OAS/RNaseL系统和RNA聚合酶Ⅲ介导的抗病毒通路,还能通过抑制ISG15的修饰活性、ADAR1催化腺苷酸转化为次黄嘌呤的编辑活性以及IRF3/7的功能进一步逃避宿主的免疫监控。本文就E3蛋白介导的抗干扰素免疫逃避策略做一简要综述。 Vaccinia virus(VV) can antagonize the anti-interferon(IFN) action of host,which is an important mechanism of VV pathogenesis.E3 protein is a key immune evasion factor encoded by VV,which functions by inhibiting the antiviral pathways mediated by PKR,2′,5′-OAS/RNaseL system and RNA polymeraseⅢ.On the other hand,E3 can also evade the host immunosurveillance through inhibiting the modification activity of ISG15,the A-to-I RNA editing activity of ADAR1 and the activation of IRF3/7.This review focuses on introducing the anti-IFN evasion strategies mediated by E3.
