血管内皮生长因子对血管内皮细胞ADAMTS5、ADAMTS4及MMP-9表达的影响 被引量:6

摘要 目的探讨不同浓度血管内皮生长因子(VEGF)刺激人脐静脉血管内皮细胞(HUVEC)24 h后ADAMTS5、ADAMTS4、MMP-9的表达。方法体外培养HUVEC,用不同浓度(0,10,20,50 ng/ml)rhVEGF165刺激24 h,以Real time PCR和Western印迹检测ADAMTS-5、ADAMTS-4、MMP-9基因和蛋白水平的表达。结果随VEGF165刺激浓度的增长,实验组(10,20,50 ng/ml)ADAMTS5、ADAMTS4、MMP-9基因及蛋白水平表达均高于对照组(0 ng/ml)(P<0.05)。结论 VEGF刺激血管内皮细胞,引起ADAMTS5、ADAMTS4、MMP-9基因及蛋白表达上调,进而有可能促进动脉粥样硬化斑块内基质降解,引起斑块破裂,提示ADAMTS5、ADAMTS4、MMP-9在动脉粥样硬化发生及进展中可能发挥重要作用。 Objective To investigate expressions of ADAMTS5,ADAMTS4,MMP-9 in human umbilical veins endothelial cells (HUVECs) stimulated by different concentration of rhVEGF165.Methods HUVECs were cultured and stimulated by different concentration of rhVEGF165 (0,10,20,50 ng/ml).Real-time PCR and Western blot were used to detect mRNA and protein expressions of ADAMTS-5,ADAMTS4,MMP-9.Results The results of Real-time PCR and Western blot indicated that expressions of ADAMTS-5,ADAMTS4,MMP-9 in experimental groups (10,20,50 ng/ml) were significantly higher than those of the control group (0 ng/ml) (P < 0.05).Conclusions VEGF could stimulate the expressions and activities of ADAMTS-5 ADAMTS4 and MMP-9 of HUVECs,they could increase the breakdown of extracellular matrix and enhance the vulnerability of atheromatous plaque,which indicates that ADAMTS5 ADAMTS4 and MMP-9 may play important role in the occurrence and development of atherosclerosis.
