摘要
利用番鸭胚从患有以软脚为主要临床症状 ,以肝、脾表面有多量白点 ,肾肿大、出血为主要病变的疫病 (暂称为“番鸭肝白点病”)的雏番鸭肝、脾中分离到 5株病毒。番鸭胚经病毒接种后 2~ 5 d死亡 ,胚体枕部、颈部、背部充血、出血 ,部分胚体肝、脾表面有灰白色小点 ,直径约 0 .5 mm。病毒经适应后能在番鸭胚成纤维细胞中增殖并传代 ,产生细胞病变。雏番鸭经人工感染后出现与自然发病鸭相同的临床症状和病理变化 ,并回收到病毒。经电镜观察 ,病毒呈球形 ,正二十面体、立体对称 ,无囊膜 ,直径为 6 0 nm左右。病毒粒子在胞浆中增殖。病毒对胰蛋白酶、氯仿和 5 0℃处理 1h不敏感 ,对紫外线、 6 0℃处理 1h、 70℃处理 1h、 80℃处理 1h敏感 ,病毒核酸型为 RNA;病毒不能致死鸡胚 ;对鸡、鸭、鸽红细胞均无凝集作用 ;与番鸭细小病毒、鹅细小病毒和鸭流行性出血症病毒无抗原交叉关系 ,表明该病毒是番鸭肝白点病的病原 ,是一种新的
s Five strains of virus were isolated from the liver and spleen of Muscovy Duck with sym ptoms of week foot, lots of white spots in liver and spleen,swelling and hemor- rhage of kidney on Muscovy Duck farms in Fujian during 1997.Muscovy Duck em bryo inoculated with the isolated strain died 2 - 5 days with congestion and homorrhage,and sometimes with lots of white spots about 0 .5 mm in diameter in liver and spleen.The isolated virus could be propagated in Muscovy Duck em bryo fibroblastcell cultures with cytopathic effects.Muscovy Ducks inoculated with the isolated virus showed pathology similar to thatof field cases,and the isolat- ed virus recovered from the tissues of these ducks.The isolat- ed virus showed spherical,icosahedral smmetry,no envelope, RNA- containing particle,about 6 0 nm in diameter,inagglu- table red cells of chicken,duck and pigeon,no pathogentic to chicken cmbryo.The virus replication occured in cytoplasm. It was resistant to treatment with chloroform,trypsin and 5 6℃ (1h) ,sensitive to ultraviolet rays,6 0℃ (1h)、 70℃ (1h) and80℃ (1h) .There were no antigen correlation be- tween the isolated virus and MPV or GPV by IFA,between the isolated virus and DEHV by AGP.The results showed that the isolated virus is the pathogen of the new infectious disease of Muscovy Duck Characterized by week foot,lots of white spots in liver and spleen,swelling and hemorrhage of kidney and is a new member of RNA virus.
