实时定量PCR检测VEGFs在宫颈癌的表达以及与淋巴结转移的关系 被引量:3

Expression of Vascular Endothelial Growth Factor in Cervical Cancer and Its Relationship with Lymph Node Metastasis

摘要 目的研究血管内皮细胞生长因子(vascular endothelial growth factor,VEGF)A,C,D以及淋巴管内皮细胞标记物Prox-1在宫颈癌组织中的表达情况,分析其与宫颈癌临床分期和淋巴转移的关系,以及VEGF-C/VEGF-D/VEGFR-3信号途径之间的相互关系。方法采用逆转录酶-聚合酶链反应(reverse transcri ptase-polymerase chain,RTPCR)及Real-time RT-PCR法分析100例宫颈癌组织和20例正常宫颈组织中VEGF-A,C,D以及Prox-1的表达情况。结果 VEGF-A,D和Prox-1的表达水平在淋巴转移组织和未发生淋巴转移的组织中有统计学差异,VEGF-C在癌症组织的表达低于正常组织,但随着临床分期的增加,呈一定的增高趋势。结论 VEGF-A,C,D在淋巴结转移过程中发挥了重要作用,可能从多方面影响宫颈癌的发生、发展和转移。 Objective To study the expression of vascular endothelial growth factor (VEGF) A, C, D and lymphatic endothelial cell marker Prowl in cervical cancer, analyze their relationship with the clinical stage of cervical cancer and lymphatic metastasis, and interactions among VEGF-C/VEGF-D/VEGFR-3 signaling pathways. Methods Reverse transcriptase-polymerase chain (RT-PCR) and real-time RT-PCR was performed to test VEGF-A, C, D, and Prowl expressions in 100 cervical cancer tissues and 20 normal cervical tissues. Results VEGF-A, D and Prox-1 expressions were significantly different in lymph node tissues with or without lymph node metastasis. VEGF-C expression in cancer tissue was lower than that in normal tissue. Conclusion VEGF-A, C, D play the important roles in the lymphatic metastasis and possibly from multiple aspects impact the cervical cancer development and metastasis.
