口服低剂量硝酸镧对糖尿病大鼠心肌NF-κB和TNF-α表达的影响

摘要 目的:观察口服低剂量硝酸镧对糖尿病大鼠心肌NF-κB、TNF-α表达的影响。方法:以Wistar雄性大鼠45只为研究对象,取35只造模,造模成功后,将实验动物分为3组即对照组(10只)、糖尿病模型组(18只)和硝酸镧给药组(12只),3组大鼠分别每日给予生理盐水、生理盐水、硝酸镧(0.2 mg/kg)灌胃1个月,然后处死取血液和心脏,采用放免法检测大鼠血浆中血糖、甘油三酯(TG)、总胆固醇(TC)、NF-κB及TNF-α的含量;RT-PCR法检测大鼠心肌组织中NF-κB及TNF-α的mRNA表达水平;免疫组化法检测大鼠心肌组织中NF-κB及TNF-α的蛋白表达;HE染色光镜下观察心肌结构。结果:糖尿病模型组大鼠血浆中血糖、甘油三酯(TG)、总胆固醇(TC)、NF-κB及TNF-α的含量明显高于对照组和硝酸镧给药组且大鼠心肌组织中NF-κB及TNF-α的mRNA水平和NF-κB及TNF-α的蛋白水平也明显高于对照组和硝酸镧给药组;光镜下可见对照组大鼠心肌细胞排列紧密整齐、糖尿病组大鼠心肌细胞排列疏散紊乱间质可见炎细胞浸润、硝酸镧组大鼠心肌细胞基本整齐紧密。结论:口服硝酸镧(0.2 mg/kg)可抑制糖尿病大鼠NF-κB、TNF-α的表达对糖尿病大鼠心肌组织有一定的保护作用。 Objective:To observe the effects of lanthanum nitrate low-dose oral on diabetic rat myocardial NF-κB and TNF-α expression.Methods:45 male Wistar rats were studied,35 of the model,after the success of the model,the experimental animals were divided into three groups namely the control group(10),diabetic model group(18) and La(NO 3) 3 treatment group(12),Group 3 rats were given daily saline,daily saline,La(NO 3) 3(0.2 mg/kg) orally one month,and then were executed blood and heart.By radioimmunoassay in rat plasma glucose,triglyceride(TG),total cholesterol(TC),NF-κB and TNF-α content.RT-PCR assay the rat myocardium NF-κB and TNF-α mRNA expression.Immunohistochemical method to detect the protein expression of NF-κB and TNF-α in the rat myocardium.HE staining observed myocardium structure.Results:Diabetic model rats plasma glucose,triglyceride(TG),total cholesterol(TC),NF-κB and TNF-α content was significantly higher than the control and administration of lanthanum nitrate group and rat myocardium NF-κB and TNF-α mRNA levels and NF-κB and TNF-α protein levels were also significantly higher than that in the group and lanthanum nitrate dose group.Light microscopy showed the control group rat myocardium cells tightly packed and tidy,diabetic rats myocardium cells arranged evacuation disorder interstitial visible inflammatory cell infiltration,the lanthanum nitrate group rat cadiomyocytes basic tidy close.Conclusion:Oral administration of lanthanum nitrate inhibits diabetic rat NF-κB and TNF-α expression have a protective effect on diabetic rat heart.
