慢性牙周炎吸烟患者外周血单核细胞TLR受体的表达及意义 被引量:1

Expression and Significance of Toll-like receptors on CDl4 + Mononuclear Cells from patients with Chronic Periodontitis of Smoking

摘要 目的探讨慢性牙周炎吸烟患者外周血CDl4+单核细胞TLR2和TLR4的表达及意义。方法选择慢性牙周炎吸烟患者(PS组)20例、慢性牙周炎非吸烟患者(PNS组)31例及健康不吸烟者20例(NS组),应用流式细胞仪(FCM)全血双色免疫荧光直标法检测外周血单核细胞的TLR2和TLR4表达水平,结果以CDl4+细胞TLR相对平均荧光强度(mfi)和TLR相对阳性细胞百分率(rpcp)表示,对各组进行对比研究。结果①慢性牙周炎吸烟患者(PS组)CDl4+单核细胞TLR2表达的mfi及rpcp均明显低于慢性牙周炎非吸烟患者组(PNS组)和健康组(NS组);PNS组TLR2的mfi明显低于NS组,而rpcp与NS组相比差异无统计学意义。②PS组CDl4+单核细胞TLR4表达的mfi明显低于PNS组,与NS组相比差异无统计学意义,而rpcp低于PNS组和NS组;PNS组TLR4表达的mfi明显低于NS组,而rpcp与其相比无统计学意义。结论慢性牙周炎吸烟患者外周血单核细胞TLR表达显著降低,其先天性免疫可能受到抑制。 Objective To investigate the expression of toll-like receptor-2 ( TLR2 ) and toll-ike receptor-4 ( TLR4 ), on CD14 + mon- ocytes of patients with chronic periodontitis of smoking, and to explore the role of TLR2 in patients with chronic periodontitis of smok- ing. Methods Choose 20 patients with chronic periodontitis of smoking( PS ) , 31 patients with chronic periodontitis without smoking ( PNS ) , and 20 healthy non smokers ( NS ) . The expression of TLR2 surface molecules on human CD14 + monocytes was assessed using nuorescence activated cell sorter allalysis by flow cytometry, expressed as relative mean fluorescence intensity ( mfi ) and relative posi- tive cell percent ( rpcp ). Results The mfi and rpcp of TLR2 on CD14 + monocytes of PS were both significantly lower than PNS and NS. The mfi of TLR2 of PNS was lower than NS, the rpcp of PNS was similar to NS. The mfi of TLR4 on CDI4 + monocytes of PS were significantly lower than PNS , but it is similar to NS . The rpcp of TLR4 of PS were significantly lower than PNS and NS. The mfi of TLR4 of PNS was lower than NS, the rpcp of PNS was similar to NS. Conclusion The expression levels of TLR2 and TLR4 on CD14 + monocytes of patients with chronic periodontitis of smoking decreased significantly. The innate immune response may be depressed in those patients.
