高糖对肾小球系膜细胞增殖的影响

摘要 目的探讨盐皮质激素受体(MR)和大分子丝裂原活化蛋白激酶(BMK1)激活在高糖诱导肾小球系膜细胞增殖中的作用。方法使用[1H]同位素标记的胸苷结合DNA评估细胞增殖。高糖刺激肾小球系膜细胞36h后评估细胞增殖。15.5mmol/L葡萄糖刺激肾小球系膜细胞0—60min后,蛋白印迹分析BMKl表达。依普利酮(0、1、10、100μmol/L)预先处理肾小球系膜细胞60min,高糖刺激肾小球系膜细胞30min,利用蛋白印迹分析检测磷酸化的BMKl表达变化。结果15.5mmol/L高糖刺激36h能明显提高[^3H]同位素标记的胸苷表达[(1.44±0.15)倍]。10μmol/L依普利酮预先处理能明显抑制高糖刺激引起的[^3H]同位素标记的胸苷过高表达[(1.07±0.21)倍]。高糖刺激肾小球系膜细胞0—60min,在5~30min磷酸化的BMKl明显增加,激活的BMKl转移到细胞核明显增加。10、100μmol/L依普利酮浓度依赖性抑制了高糖引起的BMKl活化。结论高糖引起肾小球系膜细胞增殖和BMKl磷酸化,盐皮质激素受体拮抗剂依普利酮浓度依赖的抑制了高糖引起的BMKl活化以及肾小球系膜细胞的增殖。
