摘要
目的观察淫羊藿黄酮和淫羊藿素对人骨肉瘤细胞株Saos2以及缺氧诱导因子(HIF1α)、Caspase-3和Bcl-2基因表达水平影响。方法 CCK-8法检测各种浓度对Soas2细胞增殖的影响;倒置相差显微镜观察作用后肿瘤细胞的形态学变化;AnnexinV/PI双标记法流式细胞术检测淫羊藿素诱导Saos2细胞早期凋亡的情况;RT-PCR半定量检测HIF1α、Caspase-3、Bcl-2的表达量。结果淫羊藿总黄酮和淫羊藿素都可抑制Soas2细胞增殖,并可致早期凋亡,RT-PCR半定量检测到HIF1α、Bcl-2的表达量降低而Caspase-3增高。结论淫羊藿素能抑制骨肉瘤Soas2细胞的增殖与HIF1α表达下调相关,诱导其产生早期凋亡,与Caspase-3的高表达及Bcl-2表达下调有关。
AIM To observe the effect of total flavonoids from Epimedii Folium and icatitin on osteosarcoma cell line Saos2 in vitro and the HIF1 or, Caspase 3, Bcl-2 gene expression level. METHODS CCK-8 method was used to test the effects of proliferation inhibition. The inverted phase contrast microscope was used to observe morphological changes of Saos2. AnnexinV/PI double marker method and flow cytometry were used to detect the early Saos2 cells apoptosis induced by icatitin. RT-PCR was used to detect the HIF1 ct, Caspase-3 and Bcl-2 gene expression level. RESULTS Total flavonoids from Epimedii Folium and icatitin could both inhibit the proliferation of Saos2, and induce Saos2 cells apoptosis. Icaritin could down-regulate HIF1α, Bcl-2 and upregulate the Caspase-3 gene expression level. CONCLUSION Iearitin can inhibit Soas2 cells' proliferation in relation to down-regulation of HIF1α, induce the early apoptosis in association with up-regulation Caspase-3 and decrease Bcl-2 gene expression level.
