摘要
针对胶原水凝胶力学性能及成形性能差的问题,采用琼脂糖与胶原复合的方法制备了一种兼备良好生物学、力学及成形性能的琼脂糖/胶原复合水凝胶材料。研究了胶原与琼脂糖溶液的复合方式,制备了具有相同胶原质量浓度、不同琼脂糖质量浓度的琼脂糖/胶原复合水凝胶。以胶原水凝胶为对照组,从力学强度、微结构成形性和细胞相容性方面综合评价了不同琼脂糖/胶原复合水凝胶材料的性能。结果表明:随着琼脂糖浓度的提高,复合水凝胶的力学性能成倍增加,微结构的成形性能也逐步改善,但细胞相容性却逐渐降低。琼脂糖质量浓度为0.004g/mL、胶原质量浓度为0.003g/mL的复合水凝胶能较好,兼顾了水凝胶的力学性能、成形性及细胞性能。体内植入实验发现,该配比的复合水凝胶在体内能有效减缓胶原水凝胶的降解,并产生较低的免疫排斥反应。研究的琼脂糖/胶原复合水凝胶有望用于构建具有仿生血管系统的组织工程支架。
Aiming at the poor mechanical property and formability of hydrogels,agarose is incorporated with collagen to prepare a composite hydrogel with good biological,mechanical and structural properties.The blending methods of agarose and collagen solutions are investigated,and the composite hydrogel solutions with same collagen content and different agarose contents are finally prepared.The comprehensive properties of the resultant hydrogels derived from different composite solutions are evaluated and compared with collagen hydrogel.The results indicate that with the increase of agarose content,the mechanical property and microstructure replication property of the composite hydrogels are gradually improved while the biological property decreases.The hydrogel with 0.003 g/mL collagen and 0.004 g/mL agarose is chosen as the optimal agarose/collagen composition.In-vivo implantation experiment verifies that such agarose/collagen composite hydrogel effectively weakens material degradation and simultaneously induce moderate rejection response.This agarose/collagen hydrogel is expected to be used for fabrication of organ scaffolds with biomimetic vascular system.
