猪PLIN1基因单核苷酸多态检测与遗传分析 被引量:1

Single nucleotide polymorphism detection and genetic analysis of porcine PLIN1 gene

摘要 以皖南黑猪、安庆六白猪、圩猪和霍寿黑猪共236头猪为研究对象,采用PCR-SSCP方法检测猪脂滴包被蛋白(Perilipin,PLIN1)基因的遗传多态性。结果表明,在皖南黑猪、安庆六白猪PLIN1基因3 519 bp碱基处发生A>G突变,检测到AA、AB、BB 3种基因型,其中AA基因型频率在这2个群体中分别为8.97%和29.23%,AB基因型频率分别为43.59%和56.92%,BB基因型频率分别为47.44%和13.85%;皖南黑猪、安庆六白猪的多态信息含量(PIC)分别为0.335 3和0.369 0,在该位点均处于中度多态;卡方检验表明皖南黑猪群体和安庆六白猪群体均处于哈迪-温伯格平衡状态(P>0.05);在圩猪、霍寿黑猪群体中PLIN1基因的第2外显子没有检测到突变。PLIN1基因的第3外显子在4个群体中均没有检测到突变。 Two hundred and thirty-six pigs from Wannan Black pig, Anqing Liubai pig, Wei Pig and Huoshou Black pig populations were used for detecting the polymorphism of PLIN1 gene by PCR-SSCP method. The results showed that a A〉G mutation at 3 519 base position, and 3 genotypes (AA, AB and BB) were found in Wannan Black and Anqing Liubai. The genotypic frequency of AA was 8.97%, 29.23%, respectively, AB was 43.59%, 56.92%, respectively, and BB was 47.44%, 13.85%, respectively. The polymorphism information content (PIC) was 0.335 3 in Warman Black population and 0.369 0 in Anqing Liubai population, which were all in aver- age polymorphism situation. According to the Z2 test, there were in Hardy-Weinberg equilibrium in Wannan Black and Anqing Liubai population (P〉0.05), but there was no mutation in exon 2 in Wei Pig and Huoshou Black pig populations. Moreover, there was no mutation in exon 3 in these four populations.
